In this experiment,a single factor test was used.20 healthy 60-day-old male blue-brown chickens were randomly divided into 4 groups,5 in each group,which were 0 g/kg K2CrO7control group?CG?and 0.2 g/kg K2CrO7 low dose.Group?LG?,0.3 g/kg K2CrO7 medium dose group?MG?,0.4 g/kg K2CrO7 high dose group?HG?,the poisoning group was drinking potassium dichromate aqueous solution,the control group was routinely drinking water,and the chicken was used for 21 days,kill chicken and take tissue.Chicken tissue samples of heart,liver,kidney,spleen,muscles and serum were digested by microwave digestion.There are different levels of mass spectrometry and non-mass spectrometry interference in chicken tissue samples with high organic content and complex body composition.52Cr is selected as the element to be tested,and the kinetic energy discrimination mode KED in the collision reaction cell technology CRC is used to eliminate or reduce the mass spectrometry and non-mass spectrometry interference by using the online internal standard72Ge and optimizing the instrument parameters.A method for the determination of chromium in chicken tissues treated by potassium dichromate by microwave digestion-inductively coupled plasma mass spectrometry?ICP-MS?was established.The effects of different concentrations of potassium dichromate on the chromium content in heart,liver,spleen,kidney,muscle and serum of chicken tissues and the distribution of chromium in tissues were investigated.The results showed that drinking chromium-added water caused slow growth of chickens.The standard curve of the established method ranged from 2 ng/mL to 50 ng/mL.In the linear range,the correlation coefficient was R2=1,and the detection limit of the instrument was 0.0091 ng/mL,the method detection limit is 0.135 ng/mL,and the limit of quantification is 0.448 ng/mL,which meets the requirements of the determination.In the precision test?n=8?,the relative standard deviation is less than 5%,and the standard recovery of the method is 96.2%100.2%.The average value of the assay was 10.964 ng/mL and the relative standard deviation was4.83%,which indicated that the instrument was stable.The content of chromium in the national standard substance?GBW10018?of chicken meat was determined to be 0.593±0.005?g/g,which is basically consistent with the reference value of the standard substance of0.59±0.11?g/g.The test results showed that the accumulation of chromium in the chicken body was mainly the liver and kidney,followed by the spleen,heart and muscle,which accumulated the least in serum.The above-mentioned indicators are used to investigate the method,which is simple,fast,accurate and sensitive,and is suitable for analysis of large quantities of samples.The determination of chromium content in chicken tissues treated with potassium dichromate not only confirmed the distribution of chromium in chicken tissues,but also provided an auxiliary examination method for the establishment of the diagnosis of chicken chromium poisoning.Provide a basis for developing effective chromium-removing drugs.
