With the development of modern science and technology,organic fluorescent probes are widely used in various fields for detection of substances duing to their simple,rapid,sensitive and selective methods;for example,metals cation such as Cu2+,Hg2+,Fe2+,Fe3+,and Zn2+;ClO-,F-,Cl-,Br-,I-,CO32-and other anions;H2O2,NO,H2S and other small molecules.Combined with fluorescence microscopy imaging technology,it is used to monitor biological targeting molecules in living cells and their important physiological activities and pathological processes in vivo.Therefore,fluorescent probes have gradually become an indispensable tool in modern science technology and clinical medical diagnosis.In this paper,three kinds of fluorescent probes were designed and synthesized to detect active substance in vivo.The main contents of the research are listed as follows:?1?A reversible fluorescein-based fluorescent probe which exhibits high sensitivity and selectivity for Zn2+and ATP,has been designed and synthesized.The sensing process was completed via fluorescence variation induced by an opening and closing of the spiro-ring of fluorescein.Zn2+/ATP-induced fluorescent intensity shows a good linear relationship with the concentration of Zn2+/ATP in the range of 0-10?M with a detection limit of 0.1?M/0.5?M.Moreover,the probe was further applied to visualize and detect Zn2+and ATP in vitro and in vivo.?2?A highly sensitive and selective fluorescein-based fluorescent probe for the efficient detection of OCl-over other ions has been developed.The probe was successfully applied for quantitative detection of OCl-in real life water samples and bovine plasma with satisfactory results.Confocal fluorescence imaging of cells for detecting OCl-in vivo was carried,which could provide a potential and powerful approach for the detection of OCl-in biological system.?3?A H2S fluorescent probe 3 was designed and synthesized.The probe used thiophene chloride to modify the naphthyl fluorescein precursor.The target product was characterized by NMR and MS.Fluorescence detection,UV absorption detection,response time,competition and selectivity of probe 3 were tested,indicating that probe 3 is a highly sensitive and highly selective probe of detecting H2S.Probe 3 can be used as a test strip to measure H2S in water samples and as a fluorescent gel to visually recognize H2S.
