Taking high purity C-glycoside flavonoids of bamboo leaf(H-BLF)with above 90%total mass of C-glycoside flavonoids and turnip polysaccharide(TP)as raw materials,and taking ?-polylysine(?-PL)as cross-linking agent,a kind of nanoparticle(H-BLFnp)was prepared by ionic cross-linking method and vacuum freeze-drying technology.The structural and properties characterization of H-BLFnp were tested,then its skin-care effect on moisturizing,whitening and anti-aging was evaluated both in vitro and in vivo.The main contents and results are as follows.(1)The nanodrop(H-BLFnd)was prepared through ionic cross-linking method,taking H-BLF as core material,TP as packing material,and ?-PL as cross-linking agent.After orthogonal test optimization,the preparation parameters of H-BLFnd included:3 mg/mL of TP,12:1(w/w)mass ratio of TP to H-BLF,0.4 mg/mL of ?-PL and 300 mL/h of constant flow pump velocity.Then,8%(w/v)of ?-cyclodextrin(?-CD)was added to H-BLFnd as protective agent,and the nanoparticle(H-BLFnp)was obtained via vacuum freeze-drying process.H-BLFnp is a kind of white powder,smoothly and uniformly,which is of(92.39±0.34)%encapsulation efficiency,(679±10.2)nm average size and(0.349±0.004)dispersion coefficient(PDI).(2)The chemical structural characterization of H-BLFnp were tested through UV and FTIR.The UV spectra of H-BLFnp showed typical I and II absorption peaks of flavone,The FTIR spectra showed that H-BLFnd had entered y-CD cavity and formed inclusion compound.The thermal stability of H-BLFnp was evaluated through DSC and TG,results showed that compared with H-BLFnd,the melting temperature of H-BLFnp increased 7.91 ?,the heat enthalpy increased 38.36 J/g,the decomposition temperature increased from 246? to 324?.These data showed that H-BLFnp has higher thermal stability than H-BLFnd.(3)H-BLFnp's absorption rate of moisture was evaluated at constant humidity taking glycerol as positive control,and result showed that H-BLFnp has higher hygroscopicity than glycerol(p<0.05)at 36 h.Using mouse B16 melanoma cell model,the influence of H-BLFnp on relevant indicators in the process of cell culture in vitro was studied,and the whitening efficacy was evaluated taking ?-arbutin as positive control.Results showed that H-BLFnp significantly inhibited B16 cell proliferation,tyrosinase activity and melanin synthesis compared with ?-arbutin(p<0.05),the inhibition rates achieved 45.14%,72.55%and 54.01%respectively when the concentration of H-BLFnp was at 100 ?g/mL.Othermore,the test of skin and eye irritation in mice showed that H-BLFnp has no irritation to skin and mucosa membrane and is safe for external use.(4)The skin-care efficacy of H-BLFnp on moisturizing,whitening and anti-aging was evaluated on human body by testing indexes of subjects before and after using samples taking H-BLFnp as cosmeceutical.Clinic test showed that H-BLFnp can significantly increase skin moisture,skin elasticity,brightness and gloss,and reduce skin melanin content and percutaneous moisture loss.It was elucidated that H-BLFnp has the effect of replenishing water,reducing water loss,improving skin elasticity,reducing pigment and luster on the human bodyIn summary,H-BLFnp,a novel composite bionanomaterial taking H-BLF as leading material,was made of natural ingredients and by green process.It is a kind of hydrophilic,stable,safe,non-irritating cosmeceutical and has remarkable moisturizing,whitening and anti-aging effects.H-BLFnp will be of good application prospects in daily chemical products,especially in water-soluble cosmetics...
