The Detection Performance Evaluation Of Three Kinds Of Malachite Green ELISA Kit

Malachite green is a green crystal with a metallic luster,also known as the basic green and strict block green,ma lachite green,which is both a fungicide,and dye,soluble in water,the solution was blue green.Studies have shown that malachite green residues in fish for a long time,with high toxicity,high residue,as well as carcinogenic,teratogenic,mutagenic and other side effects.In May 2002,the C hinese government listed malachite green in the list of banned veterinary drugs and their compounds,prohibiting malachite green from being used in the breeding and transportation of all food animals.In order to screen ELISA kit is more effective,rapid,specific and accurate detection of malachite green residues,the detection performance of the four brands of malachite green ELISA kit was evaluated,based on the sample pretreatment methods and conditions,detection procedure,average recovery,cross reaction rate,detection limit the linear range,the analysis and comparison,so as to understand the kit performance,better monitoring of aquatic products using malachite green in the breeding and sales process,and provide the reference and related test and research work.The results show that the detection time,the brand B kit to determine the shortest time required;on the sample pre-treatment,determination of water samples in four kit were simply treated,but in the determination of tissue samples,brand B pre-processing step is the most simple,the shortest time of range and IC50 online;on the linear range of brand A kit for 0.26~4.9 ng/mL,IC50 0.83 ng/mL;the linear range of the brand B kit for 0.37~1.6 ng/mL,IC50 0.72 ng/mL;the linear range of the brand C kit for 0.31~5.4 ng/mL,IC50 0.68 ng/mL;the linear range of the brand D kit for 0.41~6.3ng/mL,IC50 for 0.98 ng/mL;in repeatability,the coefficient of variation of the difference between the four kit board and board is not obvious;the recovery rate in addition,recovery of brand A kit 75.2-87.8%,brand B kit standard The recovery rate is 96.1-105.5%,the recovery rate of the brand C kit was 72.3-86.8%,the recovery rate of the brand D kit for 64.2-76.8%;in the cross reaction,brand A,brand C and brand D kit are cross reaction phenomena to a certain extent,the brand B kit is basically no cross reaction.Can effectively avoid false positive;the positive rate in,for water samples,four kinds of kit positive rate is 100%;for tissue samples,brand B kit,the positive rate is 100%,the brand A and brand C kit,the positive rate was 50%,the positive brand D kit the detection rate is 0.Conclusion: German brand B kit has the characteristics of simple operat ion,high speed,good stability,high accuracy and high sensitivity.It is more suitable for popularization and application in practice.At present,there is still a big gap between the technical performance of the malachite green ELISA kit and the imported kits,and the R & D capability needs to be further strengthened.
