In recent years,the small-molecular fluorescent probes have been widely used in the fields of medical biology,environmental protection,chemical detection and so on due to the advantages of convenient operation,high selectivity,high sensitivity,good biocompatibility and real-time detection.In this thesis,we used Tsuji-Trost reaction and developed several fluorescent probes for highly selective and sensitive detection of CO or Pd0.The main resarch work is as follows:1.We used fluorescein and 2,7-dichlorofluorescein as fluorophores and synthesized two fluorescent probes(probes 1 and 2).Based on the Tsuji-Trost reaction,these two probes used allyl ether as a reaction site and realized the highly selective and sensitive detection of CO in the presence of Pd2+.Experiments showed that both probe 1 and 2 can rapidly and selectively detect CO under mild conditions,accompanied by obvious color changes and fluorescence enhancement.Moreover,both probe 1 and 2 can be used to detect exogenous and endogenous CO in living cells,indicating both probes have potential for biological applications.2.In order to develop a near-infrared(NIR)fluorescent probe for detection of CO,we used 1,3-dichloro-7-hydroxy-9,9-dimethylacridine-2(9H)-one(DDAO)as fluorophore and synthesized a fluorescent probe DDAO-Pd.First of all,the optical response of the probe to Pd0 was explored.Experiments showed that DDAO-Pd can rapidly and selectively recognize Pd0(<120 s)under mild conditions,giving distinct optical changes.A simple test strip with this probe was successfully made to detect Pd0 and we also successfully applied this probe to detect Pd0 in real drug samples and living cells,indicating the probe has a good application prospect for Pd detection.3.Based on the excellent property of the probe DDAO-Pd for Pd0,we further explored the application potential for NIR fluorescent detection of CO base Tsuji-Trost reaction.Experiments showed that DDAO-Pd can quickly detect CO(<5 min)in the presence of Pd2+ in PBS buffer solution under mild conditions.Moreover,we also showed that DDAO-Pd can be used to detect exogenous and endogenous CO in living cells.In view of the advantages of NIR fluorescent detection and the excellent sensing properties of DDAO-Pd,this probe holds great potential for biological applications. |
