| The molasses spentwash contains dozens of dark brown colored recalcitrant organic compounds that are not degradated to conventional treatment.Due to the complex and unstable of components, the biodegradation of this stream needs to be further study. The mechanism of decolorization of molasses spentwash was studied by Aspergillus tubingensisi F12 which was the result of screening work, try to provide a theoretical basis for the actual wastewater biological treatment. The decolorization of melanoidins by Aspergillus tubingensisi F12 were investigated firstly. The adsorption function, activity of lignin peroxidase department,H2O2-producing activity and the change of wastewater components decolored before and after were determined. The experimental results showed that:The decolorization rate achieved maximum about 58%on 8th day, reducing sugar gradually decreased, dry cell weight increased continuously, and achieved maximum about 4.49 g/L on 8th day.The maximum activity of laccase,manganese-dependent peroxidase and manganese-independent peroxidase were 22 U/L,16.49 U/L and 10 U/L,respectively;The change of activity of lignin peroxidase department and H2O2-producing activity show a certain relationship with decolorization rate. The result of gel chromatography corresponds to decolorization rate, that is coming primarily from adsorption of Aspergillus tubingensisi F12 on first and second day of progress of decolorization, later on,bleaching is given priority to degradation. The progress of decolorization by Aspergillus tubingensisi F12 should be a synergistic effect,biodegrade-able take on the main role. The decolorization of molasses spentwash by Aspergillus tubingensisi F12 were studied secondly. The decolorization rate of molasses spentwash by Aspergillus tubingensisi F12 achieved maximum about 36%on 5th day, reducing sugar gradually decreased.Dry eell weight increased continuously, and achieved maximum about 12.47 g/L on 4th day, later on,entered a srable period, mycelium stoped growing. The progress of decolorization of MSW by Aspergillus tubingensisi F12 should be a synergistic effect from adsorption function, activity of lignin peroxidase department, and H2O2-producing activity, biosorption take on the main role and accounte for 64%effect of total decolorization. The mechanism of decolorization by Aspergillus tubingensis F12 were tried to further study finally. The role of biosorption action and enzyme degradation play in the decolorization by Aspergillus tubingensisi F12 have been taken a further research to adopt adding in additive, altering culture method and replace culture medium. The experiment results verify that deeolorization of melanoidins and molasses spentwash by Asper gillustubingensisi F12 should be a synergistic effect, the decoloration rate have coreesponding to growth of strains and concentration of carbon source, whole pigment degrade by lignin peroxidase department,H2O2-producing bleach wastewater. |
