Objective:The Allium Victorialis L.(AV)is a kind of foods and a traditional Chinese medicine in China,and AV has the function of hemostatic,expectorant,analgesic and so on.Chemical liver injury is a common disease threatening many people in our society,daily prevention is particularly important to personal health.The objective of this study is to explore the protective effects of the extract of AV as a plant health product to be developed for chemical liver injury(liver injury induced by CCl4),and larify the mechanism of its auxiliary protection.To provide theoretical basis and research basis for the follow-up research and development of AV.Methods:Mice were divided into 6 groups randomly and 6 in each group.All mice were adapted to the temperature of 22±2℃,and a relative humidity of 50-60%with a 12 h dark/light cycle for 1 week,at same time they were allowed to eat water and food freely.Mice in the AV groups and silymarin group were pretreated with AV extract(100 mg/kg,200 mg/kg,300 mg/kg)or silymarin(100 mg/kg)by oral gavage for one week,respectively.One week later,to induce acute chemical liver injury,all mice except for normal group were given a single intraperitoneally injected with 10%CCl4 in oil(2 mL/kg).After injection of 16 h,the mice were sacrificed with anesthesia,after that,the serum and liver were collected for follow-up experiments.ALT and AST activities in serum were detected;The expression levels of MDA,GSH-Px and GSH-ST in liver were detected;The pathological changes of liver tissue were observed by H&E staining;The expression of MPO and p-STAT3 in liver tissue was observed by IHC;The expression level of F4/80 was observed by IFC;The levels of MPO,IL-1β,IL-1α IL-6,IL-18,IL-1R1,TNF-α and caspase-1 were detected by RT-PCR or WB;The protein and mRNA expression levels of liver fibrosis markers such α-SMA,Collagen-I and TIMP-1 were detected;The expression of caspase-3,caspase-8,caspase-9,P53,Bcl-2,Bcl-xL,Bid,p-Bid,Bcl-xS/L,PARP,cIAP1,cIAP2 proteins related to the apoptosis of liver cells were detected;The protein expression level of STAT3 and p-STAT3 was detected by western blot;Analized and mapped the signaling transduction pathway for hepatocyte injury protected by extract of AV in hepatocytes.Results:In this study,in vivo experiments in mice showed that compared with the CCl4 model group,the administration group significantly inhibited the increase of the activity of ALT and AST in the serum of mice(***p<0.001);The contents of GSH-Px and GSH-ST were increased;The content of MDA was decreased,and the expression of MPO was down regulated;The expression of F4/80 was inhibited;AV inhibited the release of inflammatory factors IL-1β,IL-lα,IL-6,IL-18,IL-1R1,TNF-α and caspase-1;The expression of protein and mRNA of α-SMA,Collagen-Ⅰ and TIMP-1 was inhibited,and the expression of Bcl-2,Bcl-xL,cIAP1,cIAP2,and PARP protein expressed and down regulation of the apoptosis of hepatocytes by caspase-3,caspase-8.,caspase-9 proteins,and proteins;The phosphorylation of STAT3 protein was controlled.Conclusions:All the experimental results showed good results,which indicated that the extract of AV had protective effects on acute liver injury induced by CCl4,and AV can be used as a functional food developed to assist in protecting chemical liver injury,and even as a raw material for the treatment of liver diseases.
