| Demethylchlortetracycline(DMCTC)is a tetracyclic antibiotic,antibacterial activity than other tetracyclic antibiotics,with long-term efficiency,high efficiency and easy to be absorbed by the characteristics of the body.In order to solve the problem of pH regulation and low titer in the production of desmethetracycline fermentation,this result has been carried out the optimization of the fermentation conditions of desmethetracycline,the use of(NH4)2SO4solution instead of ammonia In the process of fermentation of chlortetracycline,the pH was regulated and different metal ions were added to the culture medium to increase the titer of the disinimycin.The results showed that the production and accumulation of organic acids in the process of metabolism of nimicamycin were analyzed and the conclusions were drawn as follow:1.By optimizing the fermentation conditions such as the pH value,the dissolved oxygen and the age of the fermentation broth,the optimum control pH was 5.9-6.1,the oxygen control range was 30-60%and the best age was 35-52h.2.The use of 30%(NH4)2SO4 and 10%NaOH solution with the use of alternative ammonia on the desmethetracycline fermentation process to control the pH.The addition of(NH4)2SO4 solution alone is not conducive to the stability of pH in the fermentation process of methicillin,resulting in the imbalance of the internal environment and the pH instability.However,after the combination with NaOH,the pH of the fermentation process is stable(NH4)2SO4,the titer of desmethylcycline was up to 7462μg/mL,which was significantly higher than that of the control(13.5%).(NH4)2SO4 was used instead of ammonia to regulate the pH of the nematoxycin fermentation process and avoid the use of ammonia In the human body damage,reduce the occurrence of security incidents.3.The addition of Fe2+in the fermentation medium inhibited the desmethetracycline titer.The addition of 0.003%MoO42-could increase the desmethylclinexin titer of 15.6%and the final titer of 7604μg/mL.This method does not need to change the existing fermentation equipment and the original control process,and can increase production,reduce the comprehensive cost and increase economic efficiency. |
