Abstract/Summary:
It has been proved that Cordyceps militaris have important physiological function for human body.Iron element plays an important biological function in human body.To carry out the research of iron-rich C.militaris,which have great significance in theory and practice on the development of additional functions of products of C.militaris.This paper research from three aspects,such as the iron resistant properties of the iron-rich C.militarise the Condition of the submerged fermentation of the C.militaris and fermentation of the fermentor.Firstly,through the iron resistant experiment of iron-riched C.militaris,we confirmed the scope of mycelia tolerance concentration of C militarzis of iron was 0~1.6 g/L.Then,we added different concentrations of iron in the liquid fermentation,and investigated the biomass and enrichment of iron as examining index.The conclusion was that the best concentration of iron of iron-rich C.militaris was 0.06 g/L.We added concentrations of 0.06 g/L iron in the fermentation broth,and explored the relationship between the residual iron,absorption of iron and iron absorption of mycelium powder of C.militaris.The experiment showed that the iron element was rarely in the table of mycelium iron and in the compared mycelium,and less than 1%of adding iron.It is negligible.So we can confirm the iron enrichment by detecting the iron content in the mycelium powder.By examining the growth curve of the concentration of 0.06 g/L iron of iron-rich C.militaris,we have come to the conclusion that strains could reach the maximum biomass and high enrichment of iron which were cultured for 72 h.This research was on the design of single factor and multi factor orthogonal experiment,to determine the best fermentation culture medium formula of iron-rich C.militaris fermentation in shake flask for per 1 liter fermentation broth prepared adding 35 g soybean powder,30 g sucrose,1.5 g KH2PO4,1.5 g MgSO4,5*10-5 g thiamine and 0.3 g 7H2O·FeSO4.The optimum culture conditions:pH 6,temperature is set to 26℃,ferment for 72 h,5%inoculation quantity,the volume of liquid is 100/250 mL in triangle bottle.Under this condition of fermentation,we geted the data that the biomass of iron-rich C.militaris was 18.93 g/L,and the enrichment of iron content was about 16.01 mg/g.Finally,to have fermented on the best shaking flask fermentation conditions of 50 L iron-rich C.militaris in fermentation tank,the results showed that the biomass of C.militaris and the enrichment of iron content were higher than that in shaking flask fermentation,the biomass was as high as 26.31 g/L,enrichment of iron was 16.48 mg/g.At this point,the condition of fermentation tank of the iron-rich C militaris is adding 35 g soybean powder,30 g sucrose,1.5 g KH2PO4,1.5 g MgSO4,5*10-5 g thiamine and 60 g 7H2O·FeSO4 with per liter fermented liquid,with 40%fluid volume,2%inoculation quantity,the ventilation is 0.9 m3/h.The initial pH value is 6,and training under the condition of 26 ℃ for five days.Under the same condition of the shaking flask fermentation,the ventilation is 0.9 m3/h,and the fermentation time extended to five days.The conclusion lay the theoretical basis of developing a large-scale of iron-rich C.militaris.
