In recent years,cosmetic products exported from China to the European Union and the United States.were notified frequently due to the Color additive violations.It is bad for the development of our country.Whats more,it is much less conductive to health,safety and even life of consumers.At present,the research on the synthetic organic colorants that disabled in cosmetics is not enough.Our country has not yet established appropriate national standards and industry standards of colorants detection in cosmetics.So it is necessary to establish a set of quick,easy methods to determine the colorants prohibited in cosmetics.It will provide technical support to the testing of colorants and quality control of cosmetic products.The determination methods for the colorants that are prohibited by Chinese Hygienic Standard for Cosmetics(edition 2007),Regulation(EC)1223/2009 and American FDA regulation were established respectively using high performance liquid chromatography(HPLC)and ultra-performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS).The mainly contribution of this study include the following:(2)A high performance liquid chromatographic method was developed for the simultaneous determination of 10 synthetic colorants(Acid Violet 49,Pigment Red 57,Pigment Red 53:1,Acid Yellow 36,Rhodamine B,Dispersed Yellow 3,Pigment Orange 5,Sudan ?,Sudan ?,Solvent Blue 35)in cosmetics.The cosmetics were extracted sequentially by ultrasonic technique with tetrahydrofuran(TPEF),dimethyl sulfoxide(DMSO)and methanol,then centrifuged for purification.Separation was carried out on Eclipse XDB-C18 column(5 ?m,4.6×150 mm)with a gradient elution using acetonitrile and 0.02 mol/L ammonium acetate(pH value was adjusted to 4.60 with acetic acid).Diode array detector was used to determine the colorants with the wavelength ranging from 417 nm to 640 nm.Linear relationship of 10 colorants between the peak area and the mass concentration was obtained in the range of 0.5?20.0?g/mL(r>0.999).Limits of quantitation ranged from 10 to 20 mg/kg,and the average recoveries at three concentration levels ranged from 92.9%to 108.8%with the relative standard deviation 0.5%to 6.1%(n=6).(3)A solid phase extraction-ultra performance liquid chromatography-tandem mass spectrometry(SPE-UPLC-MS/MS)method was developed for the simultaneous determination of 12 synthetic colorants(including Acid Violet 49,Pigment Red 57,Pigment Red 53:1,Acid Yellow 36,Rhodamine B,Basic Violet 3,Dispersed Yellow 3,Pigment Orange 5,Sudan ?,Sudan ?,Sudan ?,Solvent Blue 35)in lip gloss,eye shadow,lipstick and other cosmetics.The cosmetics were extracted sequentially by ultrasonic technique with tetrahydrofuran(THF),dimethyl sulfoxide(DMSO)and methanol,cleaned up by HLB solid phase extraction column.The separation was performed on Acquity UPLC BEH-C18(2.1 mm×50 mm,1.7 ?m)column with gradient elution using acetonitrile-0.05%formic acid solution,then the isolates were analyzed in multiple reaction monitoring(MRM)mode and quantified by the matrix matched external standard method.The results showed that the 12 investigated colorants displayed good peak shape within 6 min.The calibration curves of 12 colorants were linear in the range of 0.5?800?g·L-1,with correlation coefficients more than 0.99.The limits of quantitation(LOQ,S/N>10)were in range of 1.4-2000 ?g·kg-1.The recoveries of 12 colorants from spiked lip gloss,eye shadow and lipstick samples ranged from 60.8%to 118.0%with relative standard deviations(RSD)of 2.3%?10.8%.(4)The assessment of matrix effect on the detection of colorants in lip gloss,eye shadow and lipstick by UPLC-MS/MS was detailed.It showed that three matrix made different effect to 12 colorants,and the matrix has the greatest suppression for the Sudan colorants.Using SPE techniques,optimization of chromatographic and mass spectrometric condition as well as blank correction matrix matched standards can reduce or eliminate the effects of matrix effect.
