Purification Of Limonene From Citrus Essential Oils And Preparation And The Skin Anti-inflammatory Research

Limonene from Citrus Peel have more advantages like wide source,multi-function,safety,xploit deeply and comprehensively utilize of limonene to renew Citrus Peel resources,is very important for economic and social significance.First,explore the better purification process and its stability of Citrus Peel essential oils of limonene.With coarse extraction of Citrus Peel essential oil as raw material,Vacuum distilling by controlling the temperature and vacuum degree,limonene content as the index,to get the best technological conditions.The results showed that the higher of the temperature and the vacuum degree,the separation effect activity will be better.The better condition for purification process was 90℃ and 30 hPa.The limonene was very stable under the conditions of light,acid and high temperature.Secondly,using the method of mechanical agitation combined with ultrasonic to make limonene emulsion,with the average particle size of emulsion as the index,and do the single factor experiment,got the best single factor was emulsifier:HLB=12,water bath temperature=60℃,the dosage of the oil phase=16%,oil/emulsifier(S0 ratio)=0.6,stirring time=15 min,ultrasonic power=750 w,ultrasonic time=8 min,based on these results of single factor experiments,selecting three significant difference factors:oil phase proportion,oil/emulsifier(S0 ratio)and mixing time,to do the response surface optimization experiment.The results of response surface optimization showed that the effect of limonene emulsion particle size from proportion of oil phase,oil/emulsifier(S0 ratio)and the mixing time was not a simple linear relationship.the best condition to get limonene of emulsion particle size corresponding with minimum response values,as follows:the oil phase ratio=17.94,oil/emulsifier=0.686,the mixing time=6.25 min,the theory of optimum particle size was 11.39.Considering the convenience of practical operation and production,the best conditions was:oil phase ratio=18.0%,oil/emulsifier ratio=0.69,the mixing time=6 min,get the best particle size was 11.49.According to the optimal conditions for preparation citrus essential oil emulsion,by measuring the particle size of emulsion,the centrifugal stability constant,zeta potential,pH,to represe the properties and stability of the emulsion.And we got the results that the average particle size is 11.49,emulsion centrifugal stability constant is 13.82%,zeta potential is-25.97,pH is 7.22.On the other hand,we compared anti-inflammatory activity of limonene ointments with emulsion.Finally,we researched xylene induced mouse ear swelling test and influencing test of blood capillary permeability of the acetic acid in mice of limonene emulsion.And the results showed that this limonene emulsion had significant anti-inflammatory effect for skin;Xylene induced mouse ear swelling test showed that limonene soft paste has better anti-inflammatory effect.In other words,the effect of the emulsion was much better than the ointments for anti-inflammatory.
