In this paper,effects of Arachidonic acid(AA),eicosapentenoic acid(EPA)and docosahexenoic acid(DHA)on the growth of RF/6A cells in the high glucose stress conditions in vitro and their mechanisms were investigated.The result of MTT assay showed that the proliferation of RF/6A cells was promoted by high concentration of D-glucose.And the proliferation of RF/6A cells was inhibited by AA,EPA and DHA in low and high concentrations.So we choose AA(60 μM and 180 μM),EPA(60μM and 160 μM)and DHA(60μM and 200μM)added to culture of RF/6A cells.Gas chromatography was used to analyze the fatty acid composition in RF/6A cells treated with D-glucose and PUFAs.The result showed that high concentrations of glucose can modulate the absorption and the metabolism of fatty acid in RF/6A cells.And the three PUFAs could be absorbed by RF/6A cells and change fatty acid composition of RF/6A.The Elisa kit was employed to determinate the express of IL-6 and VEGF in RF/6A cells treated with PUFAs.The result showed that AA promoted the expression of IL-6 and VEGF;EPA inhibited the expression of IL-6 while promote the expression of VEGF;and DHA inhibited the expression of IL-6 significantly while exert no significant impact on VEGF expression under the condition of high concentration.But in low concentration,AA promoted the production of IL-6 while inhibited expression of VEGF;EPA and DHA inhibited expression of IL-6;DHA inhibited expression of VEGF at low concentration.The results of ROS kits showed that AA and EPA promoted the production of ROS;DHA suppressed the production of ROS observably.Therefore,in three kinds of long chain polyunsaturated fatty acids,DHA exert the best effects on suppression the injury of RF/6A cells induced by D-glucose,which was worthy of further study.The hydroxypropyl-β-cyclodextrin and DHA was used as raw material and embedded object respectively,and DHA-HP--CD inclusion complex eye drops was made successfully.The results showed that HP-β-CD can effectively improve the ability of DHA penetrating cornea.Through the analysis of IR spectra,IR feature map of package is HP-β-CD IR spectrum feature instead of DHA spectrum feature,and we inferred that DHA has been successfully entrapped.The eye drops was white transparent liquid,and its value of pH was 7.31± 0.08,which indicated that the eye drops might be no irritation to eyes.
