Abalone is precious marine product with high dietary and medicinal value.Polysaccharide is an important bioactive compound in abalone.For comprehensive utilization the processing waste,polysaccharide conjugates were extracted and purified from the abalone gonad to homogeneity,and the anti-inflammatory,antioxidant and cholesterol-lowering activity were investigated.Crude abalone gonad polysaccharide(CAGP)was extracted from gonad of abalone Haliotis Discus Hannai Ino by protease hydrolysis and ethanol precipitation.CAGP was further purified with anion exchange chromatography on a DEAE-52 colunm,and three polysaccharide fractions were obtained,named AGP-11,AGP-12 and AGP-13.The protein content of the three components was determined to be 7.06%,9.36%,8.68%,respectively.The Polysaccharide content were 52.29%,57.24%,57.52%,the sulfate radical content were 0.40%10.22%,9.99%,the alduronic acid content were 7.31%,12.59%,14.85%.The molecular masses of AGP-11,AGP-12 and AGP-13 were 8kDa,22 kDa,64 kDa.The hree components shared the same monosaccharide as rhamnose(Rha),mannose(Man),galacitiol(Gla)and glucose(Glc),the molar ratio for AGP-11 was Rha:Man:Gla:Glc=5.8:8.3:1.0:8.0,the molar ratio for AGP-12 was Man:Rha:Glc=1.0:10.0:6.4,the molar ratio for AGP-13 was Rha:Man:Glc=1.1:1.3:1.In vivo experimental results indicated that AGP-13 could inhibit dimethylbenzene induced edema in mouse ear(p<0.01)and albumen induced voix pedis edema(p<0.01).AGP-13 was also effective in inhibiting vascular permeability induced by acetic acid in mice,which was administered with 200mg/kg AGP-13(p<0.05).It is suggested that AGP-13 was an effective in inhibiting acute inflammation induced by heterogeneous antigen,such as inhibiting.Antioxidant activity of AGP-13 was also evaluated in vitro and in vivo.Experimental results indicated that AGP-13 showed notable scavenging activity on hydroxyl and DPPH radicals,with moderate reducing power.The SOD activity and T-AOC were significantly increased in the mice administered with 50 or 200 mg/kg of AGP-13(p<0.01).MDA content in blood and liver was also significantly reduced(p<0.01).It was suggested that AGP-13 possess strong antioxidant capacity.The antioxidant effect of CAGP was also invstigated on hyperlipidemia model rat.Data showed that SOD activity and T-AOC were significantly decreased in hyperlipidemia rat,and MDA content was raised(p<0.01).CAGP could effectively improve the SOD activity and TOC,and reduce the MDA content(p<0.01).It was suggested that CAGP could increase the antioxidant activity of the hyperlipidemia model rat.
