And launched Kitasamycin Kitasamycin,LiCl-UV experiments of this thesis mainly consists of three parts,one is the use of composite mutagenesis to obtain high yield strains;the second is the high yield strain as the starting strain,from the fermentation process control optimization,secondary metabolic regulation of rare earth elements on kanamycin fermentation of lincomycin fermentation process of metabolic regulation optimization;three is in the laboratory to obtain the best process based on,combined with the actual production,selection of high yielding strains used,oxygen supply and demand control,feeding,quality assurance of four aspects of the 50 L fermentor 50m3 fermentation tank test and scale-up experiment.In the end,to give full play to the strain with high yield of production capacity,improve the level of fermentation,the new profit.1)The experiment was using LiCl-UV breeding method to screen the best strain which were provided by the company living creatures.Through the orthogonal experiment design,the preliminary screening of Zn1301 was chosen as the starting strain of the nest research.By studying the effect of UV irradiation time on the positive mutation rate and mortality rate,we get the best time of UV irradiation was 90s;through studying the influence of the concentration of LiCl and Kitasamycin titer experiment,the best concentration of LiCl was 1.5%.2)Through the Kitasamycin mildew ferment process control optimization and regulation of secondary metabolism research that a convenient control technology system: Kitasamycin mildew in ferment process to take leave with adding oxalic acid and soybean oil,can solve the chaos in the fermentation broth and promote the soybean oil smooth use,and thus the stability of production;50m3 fermentation tank below the original width four leaf propeller type stirring replacement for BTD six blade radial agitator and increased diameter 50 mm,dissolved oxygen in fermentation tank are significantly improved,economic and technical indicators has also been significantly improved.3)For the first time,we tried to add the rare earth element to the fermentation medium,investigating its effect on the fermentation of its yield.The experiments proved that the addition of CS and Yt to the fermentation process played an inhibitory effect,and adding EU and La played a catalytic role on the fermentation of Kitasamycin.Through screening experiment,we got the optimum concentration of EU was 21~24mg/l,and the optimum concentration of La was 6~8mg/l.4)Through the study of fatty acids and calcium carbonate,revised the quality standard of soybean oil and calcium carbonate,put an end to the resulting abnormal production,the average annual yield increased 11879 billion.Agitator is improved,especially the application of high-yield mutant,2015 annual production capacity is greatly increased,compared with the previous two years average increase 10.34%,about saving cost 11.6 million yuan.
