| The quality of environment is basic for protecting human health from diseases.The number of deaths amounts to 5000 every year just in American and 70% of which are attributed to Pathogenic bacteria.So bacteria detection is of vital importance.In this assay,we detect bacteria successfully and realize cell imaging by use of conjugated polymers.1.We design a complex of polythiophene and prophyin,It happens efficient fluorescence resonance energy transfer(FRET)from polythiophene to prophyin.Upon the addition of bacteria,the cationic TMP is adsorbed tightly to the negatively charged membrane surface of bacteria through electrostatic interactions,making prophyin reside in much farther proximity to polythiophene.Therefore,much weaker FRET is observed,which makes bacteria dection come true.Interestingly,the method can be used to detect bacteria in tap-water,which is of more important significance.2.A new cationic poly(phenylene vinylene)derivative(PPV)is synthesized and it is used to distinguish different bacteria.Due to the negatively charged surface of bacteria,it binds to cationic cojugated PPV easily by electrostatic interation and hydrophobic interaction.The bind interaction is affected by ion stregth.Owing to the different structure and component of cell wall between gram positive bacteria and gram negative bacteria,By varying the ion strengths of the PBS buffer,single PPV molecule can discriminate gram-positive bacteria and gram-negative bacteria in a rapid and simple manner.3.We recovery and culture mammary epithelial cells and mammary tumor cells successfully.The synthetic PPV can bind to the anionic surface of cell by electrostatic interation and hydrophobic interaction,which can be observed by fluorescence microscope. |
