Preparation Of Camellia Oil For Injection Purpose

Camellia oil is a natural,healthful edible vegetable oil and therefore they are widely used in medcicine and other fields.Using camellia oil as raw material in this study,acid value and peroxide value of camellia oil were reduced by molecular distillation process and alkali deacidification process to reached requirement for injection oil.The main points and contents are summarized as follows:(1)The main quality indexes of Camellia oil were as follows:absorbance(270nm)0.038,acid value 0.338 mgKOH/g,peroxide value 3.418 mmol/kg,saponification value 188.220 mgKOH/g,unsaponification value 0.527%.Acid value and peroxide value of Camellia oil did not reached requirements of oil for injection.By gas chromatography-mass spectrometry(GC-MS),it is found that the higher components of Camellia oil is a-amyrin and lanosterol.Physiologically active compounds of Camellia oil were quantitatively analyzed.Tocopherol mainly composed of a-tocopherol,β-tocopherol.a-tocopherol 6.68 mg/100g,β-tocopherol 0.0028 mg/100g.Phytosterols are mainly composed of campesterol,stigmasterol and β-sitosterol.Campesterol 2.05 mg/100g,stigmasterol 10.20 mg/100g,P-sitosterol 33.44 mg/100g.Squalene 21.71 mg/100g.(2)Molecular distillation:based on single factor tests,the optimization conditions for Camellia oil deacidification and reduction of peroxide value by orthogonal test were performed.The result showed that,the optimum deacidification process conditions were distillation temperature 160℃,feed rate 0.5 mL/min,film scraping speed 200 r/min.The optimum reduction of peroxide value process conditions were distillation temperature 200℃,feed rate 0.5 mL/min,film scraping speed 200 r/min.With the above processing,the acid value of the camellia oil was 0.18 mg KOH/g and peroxide value was 0.52 mmol/kg.(3)Deacidification:based on single factor tests,the optimization conditions for Camellia oil alkaline deacidification by orthogonal test were performed.The result showed that,the optimum alkaline deacidification process conditions were alkali content 0.1%,alkali refining temperature 50℃,alkali refining time 30 min.With the above processing,the acid value of the camellia oil was 0.05 mg KOH/g and alkali refining yield of 70%.Peroxide value 0.52 mmol/kg.(4)The main quality indexes of Camellia oil for injection were as follows:absorbance(270nm)0.013,acid value 0.045 mgKOH/g,peroxide value 0.517 mmol/kg,saponification value 182.330 mgKOH/g,unsaponification value 0.302%.Camellia oil for injection contains trace amounts of unsaponifiables.Physiologically active compounds of Camellia oil were quantitatively analyzed.Tocopherol mainly composed of a-tocopherol,β-tocopherol.a-tocopherol 3.52 mg/100g,P-tocopherol 0.002 mg/100g.Phytosterols are mainly composed of campesterol,stigmasterol and β-sitosterol.Campesterol 1.38 mg/100g,stigmasterol 4.95 mg/100g,P-sitosterol 11.55 mg/100g.Squalene 5.33 mg/100g.
