Inositol is widely used in food industry,pharmaceutical industry and feed industry as kind of additive and biochemical raw material.In this study,the optimum process of adsorption and desorption of phytate had been studied,which used corn steep water that produced in the process of corn starch production as raw material.Optimized the hydrolysis of sodium phytate by comparing autoclave method,microwave chemical reactor method and phytase hydrolysis method,which improved the hydrolysis rate of sodium phytate,and establishes a method for rapid detection of inositol by HPLC(High Performance Liquid Chromatography).Optimized the factors of decolorization by ion exchange resin,and it also optimized the factors of desalting by electrodialysis,it also obtained high purity inositol by crystallization method.1.By using ion exchange method,determined 312 ion exchange resin as the best adsorption resin for phytate.Through the study of the adsorption and desorption properties of 312 resin,the optimum conditions of adsorption and desorption were determined: the concentration of phytic acid in deep corn water was 50.0mg/m L,flow rate was 5 m L/min,the PH was 3.5-4.0,the desorption solution concentration was 5% and the flow rate was 10 m L/min.In these conditions,the adsorption rate of resin was 97.27%,the desorption rate was 96.61% and the yield of sodium phytate was 93.97%.2.In this study,three methods,hydrolysis of sodium phytate by high temperature and high pressure reactor,microwave irradiation and hydrolysis of sodium phytate by enzymatic hydrolysis were compared.The optimum conditions for hydrolysis test of high temperature and high pressure reactor are as follows:hydrolysis time was 8 h,hydrolysis temperature was 160 ?,concentration of desorption solution was 20%.The qualitative analysis of the hydrolysate by HPLC shows that inositol existed in the hydrolysate.The hydrolysis reaction of sodium phytate solution by microwave irradiation was carried out.The optimum conditions of hydrolysis were determined as follows: the concentration of hydrolysis was 20%,the hydrolysis temperature was 100 ?,and the hydrolysis time was 80 min,the hydrolysis rate was more than 80%,which is superior to the hydrolysis method of high temperature reaction kettle.The hydrolysis rate of sodium phytate was 85% by enzymatic hydrolysis of 20 U / g substrate at p H 5.5,37 ? for 2-4 hours.3.D750 resin was selected for decolorization of inositol.The optimum decolorization conditions were determined as follows: p H was 2-4,room temperature was 25 ?,and the flow rate was 6 m L/min,decolorization rate was98.31%.The optimum conditions of electrodialysis desalination were determined as follows: voltage was 50 V,flow rate was 50 L / h,time was 90 min,desalination rate up to 95.71% and inositol loss rate was 4.29%.The inositol solution was treated by crystallization.Finally,the inositolpurity was above99.0% and the yield was 75.16%.
