Objective:This article aim to isolate bioactive compounds from the seeds of Psoralea Corylifolia by bioassay-guide method.In addition,we also determine the chemical structure of bioactive compounds by related spectral NMR data.We also study the some compounds on DGAT inhibitory activity in order to provide effective lead compounds.Methods:Finding the relevant literature and drawing on the extraction and separation methods.After the crude extract of the seeds of P.Corylifolia was obtained by methanol extraction,Separating or purifying the bioactive compounds that inhibit DGAT activity by a variety of chromatographic separation methods such as silica gel,ODS column,HPLC and other separation methods.Furthermore,determining the chemical structure of bioactive compounds through comparing with related data such as 1H-NMR?13C-NMR?HMBC data.Furthermore,further analyze the inhibition of DGAT by in vitro experiments.Results:Thirteen compounds were obtained from the isolation of seeds of P.Corylifolia by the bioassay-guide method.Through structural analysis and comparing with known data,compound 1-13 were determined as Brosimacutin E?1?,Corylifolin?2?,7-O-Methylluteone?3?,Bavachinin?4?,Neobavaisoflavone?5?,Corylin?6?Corylifol A?7?,2-hydroxy-?3-bakuchiol?8?,12R,13-diolbakuchiol?9?,12S,13-diolbakuchioll?10?,12-Hydroxyisobakuchioll?11?,Psoralidin?12?,Brosimacutin D?13?,respectively.Conclusion:13 compounds were isolated from the extract of the seed of P.Corylifolia and all were known compounds.Among the isolates,compounds 3 and 13 were first isolated from the seeds of Psoralea Corylifolia.Compounds 4,7-10,12-13displayed a high selective inhibitory activity on DGAT1 with IC50 values ranging from 68.3±1.5?M to 121.1±1.4?M.Compounds 11 showed a selective inhibitory activity on DGAT2 with IC50 values 137.9±1.1?M?M.
