To study the optimal reaction conditions of rutin enzymatic glycosylation reactions for cyclodextrin glycosyltransferase.Basing Rutin conversion rate as response,and to optimize main reaction conditions for single-factor and response surface method.The various factors and their interactive effecting for significantion of response value by examining,inferring the optimal reaction conditions for the reaction time 34 h,reaction pH5.5,enzyme dosage 25 U / mL,the reaction temperature 32 ℃.In this condition,the conversion rate of rutin enzymatic glycosylation is highest,conversion rate is up to 71.9%.The use of the method of vapor-liquid vaporization(W/O/W),the glycosylated rutin microspheres were prepared by PLGA.The preparation process conditions of microspheres were optimized by the combination of microsphere envelope and total load.Optimum technological parameter is: PLGA: methylene chloride = 1:20(W/V),emulsifying temperature and stirring speed of 55 ℃ respectively and 5000 r/min,glycosylation rutin: PLGA = 1:15(W/W).After optimizing the process parameters,the encapsulation rate of the glycosylated luding microsphere was 85.94% and the dosage was 3.78%.The microspheres of glycosylation of rutin were characterized.The size of the microsphere is uniform and the shape is round,the particle size is less than 100 mu m,and the particle size is basically in normal distribution.The quality control system of the glycosylated rutin microsphere was established.The dosage,cumulative release degree and microsphericity distribution of glycosylated reed microspheres were determined.The maximum dosage of microspheres was 3.8%,ranging from 1.6 to 3.8%.Under the optimum conditions,the dosage of microsphere was stable especially.The microspheres have small diameter and narrow distribution.The content of luding in the microsphere was measured and the average value of the experiment was 84.09%.In vitro properties of glycosylated rutin microspheres were preliminarily studied.Including: suspension of microspheres,aseptic detection,moisture content,PLGA property viscosity,microsphere stability and heat source detection.The suspension state of the microsphere is basically stable,and the state does not change with time.
