The essay using Acer truncatum Bunge seed as materia and orthogonal test and response surface test as test methods study the effects of aqueous enzymatic extraction conditions and different extraction methods on extraction rate,fatty acid composition and physicochemical properties of A.truncatum oil.The main findings are as follows:(1)The optimum conditions for extracting A.truncatum oil by aqueous enzymatic extraction with alkaline protease as catalytic preparation are as follows:(1)enzyme dosage3.5%,pH10.5,enzymatic hydrolysis time 7h,temperature 55℃,tsolid-liquid ratio 1: 4.Under this condition,the oil yield reached 51.78%.And the order of the influence of each enzymatic hydrolysis factor on the oil yield was as follows: tsolid-liquid ratio>enzymatic hydrolysis time>pH>enzymatic hydrolysis temperature>enzyme dosage. The optimum conditions for extracting A.truncatum oil with mixed enzyme(the ratio of hemicellulase to neutral protease is 2:1)as catalytic preparation are as follows:enzyme dosage 5%,enzymatic hydrolysis time 5h,temperature50℃,tsolid-liquid ratio 1:4.Under this condition,the oil yield reached 50.79%.The order of the degree of influence of each enzymatic hydrolysis factor on the oil yield was as follows: enzyme dosage> tsolid-liquid ratio>enzymatic hydrolysis time>enzymatic hydrolysis temperature.(2)the efficiency of extracting maple oil by mechanical compression,Soxhlet extraction,aqueous enzymatic extraction(single enzyme)and aqueous enzymatic extraction(double enzyme)is different,and the difference is significant.The order of oil yield from high to low is: Soxhlet extraction,mechanical compression,aqueous enzymatic extraction(single enzyme method),aqueous enzymatic extraction(double enzyme method).(3)the fatty acid components of A.truncatum oil extracted by mechanical compression,Soxhlet extraction,aqueous enzymatic extraction(single enzyme)and aqueous enzymatic extraction(double enzyme)are basically the same,mainly oleic acid,linoleic acid 11-eicosenoic acid,erucic acid and nervonic acid.The unsaturated fatty acid content accounted for more than 89% of the total fatty acids.(4)There are different in the A.truncatum oil by mechanical compression,Soxhlet extraction,aqueous enzymatic extraction(single enzyme)and aqueous enzymatic extraction(double enzyme).The oleic acid value and peroxide value of A.truncatum oil were the lowest by aqueous enzymatic extraction,the smell is the same as A.truncatum oil,without impurity,and the comprehensive quality is the best. |
