In order to improve the comprehensive utilization value of pleurotus eryngii,the enzymolysis method and Box-Behnken response surface design were used to optimize the proteolytic processing conditions of Pleurotus eryngii,and various in vitro antioxidant systems were applied.The antibacterial and anti-fatigue abilities of the peptides were evaluated by antibacterial and animal experiments.The oral liquids of pleurotus eryngii Peptide,which has good palatability and certain biological activity were developed.The main conclusions of the study are as follows:?1?The polypeptide of pleurotus eryngii was prepared by protease hydrolysis.According to the results of single factor test,the regression model between the polypeptide content of pleurotus eryngii and each single factor was analyzed and established.The optimum enzymolysis conditions for the polypeptide of Pleurotus eryngii were:The pH of the solution was 9.0,the enzymolysis time was 2.1 h,the enzymolysis temperature was 50°C,the enzyme dosage was 4000 U/g,and the polypeptide content was 0.630 mg/m L.Under the optimal conditions,the polypeptide content was 0.618 mg/m L,and the measured value reached98.09%of the predicted value.?2?The in vitro antioxidation of pleurotus eryngii with different molecular weights showed that Pleurotus eryngii has a certain ability to scavenge free radicals and reduce Fe3+.The molecular weight of 3000 to 5000 Da,which is the component of pleurotus eryngii,has the strongest ability to scavenge free radicals.Within the experimental concentration range,the clearance rate was positively correlated with the polypeptide content.With the increase of the content,DPPH·,ABTS+·,·OH and the reduction rate of Fe3+gradually increased.?3?There were significant differences in the antibacterial activities of pleurotus eryngii with different molecular weights.The polypeptide has a promoting effect on the growth of lactic acid bacteria,saccharomyces sinusoids,and the growth promoting effect of the components having a molecular weight of less than 3000 Da on the saccharomyces sinusoids is significantly higher than the components having a molecular weight of 3000-5000 Da.The polypeptide component of pleurotus eryngii with a molecular weight of less than 3000 Da has a significant inhibitory effect on Staphylococcus aureus.Animal experiments showed that pleurotus eryngii polypeptide can significantly prolong the swimming and climbing time of mice,significantly reduce the content of serum urea nitrogen,and have a significant anti-fatigue effect.pleurotus eryngii polypeptide has certain hepatic and muscular health effects,can enhance its liver function,increase liver cell nuclear clarity and denseness of liver tissue structure,and can improve muscle tissue structure and make muscle texture and lines more clear and regular.?4?Optimization of the formula of the pleurotus eryngii polypeptide oral solution:the amount of the polypeptide solution is 40%,honey is 5%,sorbitol is 0.15%,and the compound stabilizer xanthan gum is 0.2%,CMC-Na is 0.2%and carrageenan is 0.3%.pleurotus eryngii polypeptide oral liquid has good stability,fresh taste and good palatability.
