| Skin trauma is a common case in the field of clinical medicine and cosmetic plastic surgery.The design and development of biomedical functional dressings with wound repair is becoming a key issue and one of the important ways to solve the problem of wound repair.The traditional wound dressing easily caused secondary injury to the wound in the process of wound repair and was infected bacteria,which resulted in the slow healing of the wound.Hydrogel has a certain moisturizing property and convenient loading of active substances favorable to rapid wound healing.In this paper,hydrogels with antibacterial and bioactivity agents were prepared by Polyvinyl alcohol(PVA)as the basic material,compositing with chitosan(CS)with antimicrobial properties and collagen(COL)needed in the healing process of skin spraying on the surface of hydrogel.The animal models of superficial skin trauma with different trauma depth were established by laser beam,and the composite hydrogel was evaluated by repairing these superficial wounds.The feasibility of the functional hydrogel dressing designed for superficial wound repair was proved.PVA was composited with chitosan as antimicrobial agent and collagen as active component of skin repair for forming composite hydrogels(PVA-2%CS、PVA-5%CS、PVA-10%CS)with different proportions by freeze-thawing method.By testing its mechanical properties,moisture retention,antimicrobial properties,protein release and cytotoxicity,a composite hydrogel dressing PVA-5%CS was obtained with the most favorable physical and biological properties.Collagen was sprayed on the surface of the composite PVA-5%CS gel during the freeze-thawing cycle to prepare the PVA-5%CS-COL hydrogel.The animal models of shallow I,II and deep II trauma were successfully established by laser irradiation to the dorsal region of SD rats,and the general observation of trauma repair,H&E staining were performed to evaluate the effect of PVA-5%CS-COL composite hydrogel on the repair of skin trauma in different depth of SD rats.The results showed that the functional hydrogel dressing could obviously shorten the wound healing time.PVA/ CS/COL hydrogel couldpromote wound skin healing,the rapid formation of upper cortex and the reduction of scar tissue formation after wound repair better than of other hrdrogels.PVA/CS/COL has potential for applications in clinical and medical cosmetic surgery as a new functional hydrogel dressing. |
