Scallop(Patinopecten yessoensis)is an important sea-cultured shellfish species in northern China.In this paper,the glycosaminoglycan from scallop(P.yessoensis)skirt was extracted and purified.The main chemical components of scallop skirt glycosaminoglycan were analyzed.The antioxidant and hypolipidemic activities were studied.The experimental results provide important basic data for the effective development and utilization of scallop(P.yessoensis)skirt.The paper mainly includes the following parts:1.to optimize the method extraction of glycosaminoglycan from scallop(P.yessoensis)skirt(PYSG): on the base of single factor investigation,the solid-liquid ratio,enzymolysis time and enzyme dosage were selected as argument.The extraction rate of glycosaminoglycan was selected as response value.Response surface methodology was used to obtain the regression model of the relationship between the extraction rate of glycosaminoglycan from skirt of scallop and the ratio of liquid to material,enzymolysis time and enzyme dosage.The optimal conditions of extracting glycosaminoglycan by enzymatic hydrolysis were as follows: the enzyme dosage 1.44%,the ratio of liquid to material 3.94,and the enzymolysis time 2.54 h.Under this condition,the extraction rate of glycosaminoglycan reached 1.873%.2.PYSG was extracted by the optimal conditions,the removal of protein was compared by different pH values.The Sephadex G-100 dextran gel column chromatography was applied to further purify of PYSG.The content and composition of glycosaminoglycan in crude and purified samples were detected.The results showed that the total sugar content was 14.95% and 21.98%,the total protein content was 21.50% and 10.60%,the total glycosaminoglycan content was 19.40% and 63.90%,the content of sulfate group was 2.85% and 6.58 %;the content of amino galactose was 3.24%,8.72%;the content of hexuronic acid was 9.21%,14.82%;the content of aminohexose was 13.29%,27.47%.The molecular weight of the purified product was determined by high performance liquid chromatography(HPLC),which was about 19490 Da.The purified product was analyzed by infrared spectroscopy and founded that PYSG has the characteristic absorption of acidic polysaccharides.Scanning tunneling electron microscopy analysis showed that PYSG has a regular ellipsoid structure,polysaccharide molecules are not easy to happen between the conclusion.It is not easy to conclude between polysaccharide molecule of PYSG.3.The in vitro antioxidant properties of PYSG were tested by using the capacity to scavenge superoxide anion free radical ability,hydroxyl radical capacity,DPPH free radical and ABTS free radical.The results showed that PYSG had a certain degree of antioxidant activities in vitro and exhibited a dose-dependent inhibitory effect on the hydroxyl free radical,DPPH and superoxide anion free radical.When the concentration of PYSG is up to 10mg/ml,the highest scavenging rate for hydroxyl radicals reached 90.68%.At the same concentration,the highest scavenging rate for superoxide anion free radical,DPPH radical and ABTS radical reached 73.28%,84.38%,54.22%.4.The experiment method of the preventive hyperlipemia animal model were applied to investigate the hypolipidemia activity of PYSG,which was divided into three doses 100,200,400 mg/kgd,simvastatin was chosen as standard indicators.After 4 weeks the content of TC,TG,HDL,and LDL were observed.The results showed that PYSG had a significant improvement on the increase of blood lipids in mice induced by high-fat diet.The total cholesterol,triglycerides,low density lipids protein content in serum were reduced obviously in predator high fat model animal.Comparatively,the high-density lipoprotein levels in serum were promoted obviously.The experimental results showed that PYSG can promote cholesterol metabolism and prevent the deposition of lipid in arterial wall and liver,which has the effect of lowering blood fat.
