PART I PREPARATION OF THE DUAL-RESPONSE HYDROGEL AND EVALUATION OF THE HEATING EFFICIENCY AND DRUG RELEASE PROPERTY IN VITROObjective To investigate the drug delivery ability of the hydroxy propyl methyl cellulose(HPMC)/ Fe3O4/ doxorubicin hydrochloride(DOX)hydrogel in vitro.Methods Fe3O4 spheres were prepared as modified solvothermal reaction.HPMC,PVA and Fe3O4 spheres were swollen to form the hydrogel.General physical property,the scanning electron mircroscope(SEM),transmission electron microscope(TEM)were used to observe the morphology of the hydrogel.The HPMC/Fe3O4 hydrogel with different concentrations of Fe3O4 spheres were used to detect the heating efficiency in the alternative magnetic field(AMF);The ability of drug delivery under different p H circumstance and different heating time was detected.Results Fe3O4 spheres were prepared successfully.The particles dispersed uniformly in the hydrogel.The heating temperature increase as the increasing concentration of Fe3O4 spheres,indicating that the heating efficiency increases as the concentration of Fe3O4.The results of in vitro drug delivery displayed that the accumulative drug release increase in the low p H circumstance and AMF.Conclusion The prepared HPMC/Fe3O4 /DOX hydrogel could combine chemotherapy and thermal therapy.PART II UNDER ULTRASOUND: THE THERAPEUTIC EFFICIENCY OF THE DUAL-RESPONSE HPMC/Fe3O4 HYDROGEL IN VIVOObjective To investigate the ability of the therapeutic efficiency of HPMC/Fe3O4/Dox for breast tumor in vivo.Methods 4T1 breast tumor beared nude mice were used to detect the therapeutic effect.They were divided into 4 groups: NS group,Free DOX group,AMF group,and AMF with DOX group.Each group was treated with the respective treatment.After treatment,the weight and tumor volume were measured until some of them recovered to health.Results The results of therapeutic efficiency in vivo showed that no recurrence occurred in the chemo-thermal group and recurrence occurred in thermal therapy group which indicating that the HPMC/Fe3O4 /DOX hydrogel could be used for chemo-thermal therapy of tumor.Conclusion The dual-response hydrogel be used for tumor therapy and remarkably inhibited the recurrence of tumor.
