Marine fungus appeared to have adapted excellently to the marine environment,thus offering a rich source of metabolites with diverse structures and various biological activities,which has been the concern of all research fields and has the potential to be new lead structures for medicine.One of the strains with high activity was chosen for further research on its secondary metabolites.This study aims at finding potential bioactive strains and their metabolites,thus lays a foundation on exploring and utilizing new microbial pharmaceutical.The main results are as follows:1.Using different polar organic solvent to extract or soak the broth and mycelia of strains XWC21-5,XWC21-10,XWC22-4,XWC23-6,and the antibacterial and ACh E inhibitory activities of different extracts were determined by agar diffusion assay and Ellman methods,respectively.The result showed that the ethyl acetate extract part of the fermentation broth XWC21-10 had stronger antibacterial activity and ACh E inhibitory activity.2.The ethyl acetate extracts from strains XWC21-10 were separated by silica gel column using different proportion of chloroform and methanol.Eluents were collected and each 50 m L were considered as a fraction.Finally obtain 11 parts of fraction and bioassayed using the method of agar diffusion assay and Ellman.The results showed that the fraction 6,7 had the stronger antibacterial activity,while the ACh E inhibitory activity of 8,11 were outstanding.3.Optimized the detection condition on the fractions by HPLC,we found that the best condition: injection volume was 20 ?L velocity was 1 m L/min,the mobile phase ratio of methanol and water is 65:35;To judge the component of the strong activity fraction in XWC21-10 by the thin layer chromatography(TLC)technology.The characteristic color reaction of alkaloid was obviously.4.To determine the content of the total alkaloids,the determination was conducted by spectrophotometry with ultraviolet photometer,while the Huperzine A was selected as contrast substance.The results show that the linearity relationship between absorban andcontent was good when the content was 3.2~38.4?g/m L,and the regression equation is A=0.108C~0.0399,R2=0.99897(n=8),and the concentrations of alkaloid in broth and mycelium of XWC21-10 were C???=0.597 mg/m L,C???=0.576 mg/m L.It is concluded that XCW21-10 from coral fungi has antibacterial and ACh E inibitory activity,so they were deserved for further study,meanwhile provide the basis for developing the marine medicines.
