| Time-resolved fluorescence immunoassay(TRFIA)has widely used in the field of clinical medicine and life science research.The most important issue is the synthesis of rare earth chelating agent with luminescence characteristics.In order to meet the requirements of TRFIA,this paper conducts the following research:In this study,the organic intermediates was synthesized from Cbz-Lys(Boc)-OH and other compounds through reduction,oxidation,substitution,hydrogenation,hydrolysis and other reactions.Finally,the organic ligand Lys-tetraamine-H(2,2)IAM-tetrathiazolide was synthesized successfully.1HNMR,MS,IR,DSC and elemental analysis were used to characterize the structure.A new kind of rare earth bifunctional chelating agent was synthesized by connecting the organic complex with the rare earth ion Tb3+,and the luminescence properties of the chelating agent were studied.The study shows that it’s luminescence lifetime is 1168μs,the maximum excitation wavelength is 336 nm,the maximum emission wavelength is 543 nm,and the Stokes shits is 207 nm.These advantages make it possible to reduce the interference of the background luminescence in the TRFIA,and make the analysis result more accurate.It has many advantages such as strong luminescence intensity,stability and long luminescence lifetime,which meet he requirements of TRFIA. |
