Deoxynivalenol(DON),also known as vomitoxin,is a secondary metabolite that is mainly produced by Fusarium graminearum and Fusarium oxysporum.DON is widely distributed in mildewed grain crops such as wheat,barley,corn.Widespread pollution,the detection rate and the detected amount are high all over the world.DON has chemical stableility that is also difficult for toxicity of destruction in general cooking processes.DON has been considered one of the most dangerous food contaminants found in nature by WHO.International and domestic have made a limit provisions for DON in food.“ China national food safety standards of mycotoxins limit in food"(GB2761-2017)provisions of cereals and their products in the DON limit 1000μg / kg.Therefore,for the require of food safety and international trade,the detection of DON is of great significance.In this paper,the determination of immunoaffinity chromatographic purification and high performance liquid chromatography used in the determinating the content of Deoxynivalenol(DON)in wheat flour has been studied.The main determination properties such as recovery,detection limit and precision were studied.In the experiment,different extraction solvents,purification columns and leaching water were selected,then eluted with methanol and eluted on C18 column and detected by ultraviolet detector.The result is as follows:(1)The recovery rate is the highest when the amount of water for rinsing is 5mL,the extraction solvent is water and the purification column is an immunoaffinity column.(2)When the spiked concentrations ranged from 1.0μ g/mL to 10.0μ g/mL and the recoveries ranged from 89.1% to 99.7%,with 0.5μg/mL standard injection,the detection limit was 0.0179μg/mL.(3)The precision of this assay was 0.9% at 10.0 μg/mL.(4)Therefore,immunoaffinity chromatography purification-high performance liquid chromatography has the feature of low detection limit,high precision,recovery close to the true value,reproducibility,easy to operate and so on.
