| Combined photothermal therapy withchemotherapy in a platformhas received widespread attention,which is expected to replace or supplement traditional cancer treatment methods.Therefore,it is very important to design a new multifunctional drug carrier system.Gold nanoparticlesshow unique property of surface plasmon resonanceand good biocompatibility,and it issafe,nontoxic,and easy to be surface-modified,which have been used in photothermal therapy.Apoferritin is an ideal vehicle for drug delivery in chemotherapy.However,the pH of tumor tissues is 5.2,belonging toweak acidic.If rely on simple pH stimulation,only a bit of drug can be released.Therefore,the complex of apoferritin and Au nanoshell will trigger the drug releaseby laser and pH,whichresultin combination of photothermal therapy andchemotherapy.The Apoferritin@Au nanoshell was coated glucose,can be selectively endocytosis by cancer cells due to the overexpressed of glucose transporters in the tumor cells.So the synthesized composite particles have become a new multifunctional anticancer material integrating photothermal treatment,chemotherapy and recognition.In summary,the main contents of this paper are as follows:(1)Apoferritin@Au nanoshell was synthesized by electrostatic interaction and the thickness of the Au nanoshell was adjusted by controlling the growth times of Au seeds.It was proved that the plasmon resonance absorption peak is related to the thickness of the nanoshell,which the peaks gradually shift to longer wavelength with the thickness of the shell increases.The sample temperature rises under laser irradiation at the plasmon-resonant wavelength.The group with the highest photothermal conversion ability was selected for subsequent experiments from the three samples.Finally,Au nanoshell,which was coated glucose on the surface,was studied including stability,photothermal conversion efficiency,photothermal stability,cytotoxicity and cell recognition.(2)The doxorubicin hydrochloride(DOX)was encapsulated into apoferritinto achieve the synergistic effect of chemotherapy and photothermal therapy.The DOX standard curve was obtained by the absorption spectra of DOX with different concentrations.And then the drugloaded efficiency of apoferritin andthe drug release rate under different pH values were calculated.The drug release rate was greatly increased under pH/ laser stimulation after the Au nanoshell modification.The survival rate of cells was investigated under the synergistic effect of chemotherapy and photothermal therapy by cytotoxicity test.Finally,the ability of the composite particle modified with glucose was demonstrated by compared with the cell fatality rate before and after coating glucose. |
