Tumor is one of the main threats to human health.Once the tumor has deteriorated,it becomes a malignant tumor and that is cancer.At present the detection methods of tumors mainly include biopsy,nuclear magnetic resonance and CT.However,all these methods have some damage to the human body,and further deterioration of the condition can be detected if mistakes are detected.The emergence of surface-enhanced Raman spectroscopy can be a good solution to the above problems,due to SERS is a nondestructive,real-time monitoring method in vivo testing,and can achieve non-invasive detection of cancer.However,the current problems of SERS are the preparation of active substrates,including the controllable topography and size,and the control of stability and sensitivity.Therefore,the preparation of substrate materials are crucial to the development of SERS and its practical application effect.In summary,this thesis mainly focuses on the preparation of SERS substrate and the SERS detection in cancer tissues.The main content includes the following:(1)Copper-doped Au NRs were prepared by gold-copper co-blending using seed growth method.By adjusting the incorporation of copper ions and changing the ratio of gold and copper,the size and morphology of the nanorods can be adjusted to control the optical properties of the nanorods.Copper-doped nanorods were tested by UV-Vis-NIR absorption,XRD,TEM,HRTEM and other characterization methods.Finally,two kinds of nanomaterials were tested by photothermal conversion test.It was found that the temperature rise of copper-doped gold nanorods was significantly lower than that of gold nanorods within 25 min,but the overall temperature tended to rise.(2)The SERS enhancement properties of copper-doped gold nanorods were analyzed.By contrast test,the SERS performance of copper-doped gold nanorods and gold nanorods was analyzed using 4-ATP and R6G markers.Finally,The SERS properties of copper-doped gold nanorods were found to be significantly stronger than that of gold nanorods.The SERS enhancement factor of Copper-doped gold nanorods can reach 10~5.Finally,the detection limit of copper-doped gold nanorods can reach pg/mL according to the sensitivity test.The ultra-sensitive detection can be achieved.Through the stability test,it is confirmed that the doped nano-material is stable.(3)The copper-doped gold nanorods were used as active substrates to detect the lung cancer tissues.The Raman and SERS spectra of normal and cancer tissues were studied.By comparing the Raman peak position and intensity of Raman peaks,as well as mapping analysis,the cancer detection can be achieved.
