In this paper,cuttlefish skin collagen hydrolysate-gelatin and anti-oxidant peptides were prepared using rich cuttlefish skin as raw material.The preparation conditions were optimized by using single-factor experiment and response surface experiment,respectively.And study the properties of gelatin and antioxidant peptides.1.Using low concentration alkaline solution pretreated cuttlefish skin and successfully prepared the cuttlefish skin gelatin by adjusting the parameters of the main influencing factors in the preparation process,and the preparation conditions were:extraction temperature was 64.10 ?,extraction pH was 2.84,alkali pretreatment time was 1.5 h,and extraction time was 3.5 h.2.The properties of cuttlefish skin gelatin were studied in terms of basic physicochemical properties,amino acid composition,secondary structure,rheological properties and thermal stability.The results showed that the prepared substance is a nutrient-rich gelatin that reached the scope of national standards,and it contains 8.8%helix,27.1%beta-sheet,25.5%beta-turn and 37.2%random coil,respectively.It also showed excellent rheology and thermal stability.3.Alcalase was used to hydrolyze the skin of the cuttlefish and the peptides with antioxidative properties were successfully prepared by controlling the hydrolysis conditions.The hydrolysis conditions are as follows:enzymolysis temperature wss 44.48?,enzymolysis pH was 7.79,enzyme addition was 7145 u/g,and enzymolysis time was 150 min,the substrate concentration was 33.3%.4.The Antioxidant Activity of the prepared peptides was studied.The IC50 of scavenging DPPH radicals was 2.125 mg/mL,the IC50 of scavenging ABTS radicals was 1.299 mg/mL,the IC50 scavenging hydroxyl radicals was 0.7422 mg/mL,and showed a certain ferric reducing antioxidant power.We found that less than 3KD component than the 3KD component had a stronger antioxidant activity by ultrafiltration.(The IC50 is the concentration of the solution at a scavenging activity of 50%.)...
