Tea,alias tea,Cashew,Ming,etc.Tea is one of the most popular drinks in the world,but the tea consumption market is shoddy,fake or even doped fake phenomenon occurs.Depending on the traditional sensory and physical and chemical test methods,it is difficult to accurately determine the plant species doped in tea.Gene cloning sequencing detection has high sensitivity and high accuracy,therefore,this topic is aimed at the detection of tea-doped plant components by cloning sequencing of rbcL gene fragment,which can provide a new way for the detection of tea doping.In this study,37 kinds of tea samples were tested by doping plant components.A method for the qualitative detection of tea doping was carried out by using a gene sequence of plant nuclear ketone sugar-1,5-two phosphate phosphatase/Jaki(rbcL),comprises three steps: extracting total DNA from tea and amplification of rbcL gene fragment,purifying amplified product and direct sequencing;transforming purified amplified products into Escherichia coli,the positive clones were randomly selected for sequencing and sequence analysis.In the study,through the initial selection of 7 kinds of tea(Yueyang yellow tea,tieguanyin,too mu silver needle,Xinyang Maojian,Zheng Shan small species,Palace Puer,six Fort tea)sample to determine the feasibility of the method,and found that 6 of them are to some extent mixed with other plant ingredients,then 30 kinds of tea samples were tested and tested by cloning sequence of rbcL gene fragment,and 9 kinds of pu ' er tea samples were tested by doping,and other non tea plants were detected in addition to some plant ingredients reported in previous literatures.In the study,based on 37 samples of tea sample detection,in addition to detecting reports in the literature of doping components such as Glycine max,Pinus krempfii,Populus nigra,Musa etc.,were detected in plant ingredients such as Phellodendron amurense,Cucumis melo var.Flexuosus,Oryza sativa,Foeniculum vulgare and other more dopings.Except for ingredients that areeither inadvertently or unintentionally mixed,the addition of beneficial ingredients in certain reports is excluded.Because of the origin of the raw materials of tea can not be fully understood,the reasons for the existence of the composition of the plant can not be determined,and it is still to be determined in a more thorough and comprehensive study.However,it is believed that it can play a certain auxiliary role in tea safety and quality testing,and provide a possible way for tea adulteration detection.
