As known to us,the oxidation or the corruption of compound and other pollutants can make the body produces a large number of free radicals and to attack the DNA or RNA,which causes cell mutation and produces the canceration phenomenon.Oxidative damage to cells is not only associated with the onset of aging,cancer,diabetes,and many neurodegenerative diseases,but also leads to the development of cancer.So far,the antioxidative activity of licochalcone A was studied.The anticancer activity was studied using hepatocarcinoma cells as a model,which laid a theoretical foundation for the development of antioxidation,anticancer drugs and health products with licochalcone A.In this study,five different systems,including DPPH,ABTS,O2-,H2O2 and FRAP,were used to detect the antioxidant activity of licochalcone A.The results showed that the percentage of free radicals scavenging is dose-dependent,and when the concentration of licochalcone A is 64?g/mL,five kinds of detection of different systems results show the best antioxidant ability of licochalcone A.Secondly,the T-AOC and CAA methods were used to verify that licochalcone A also has a strong antioxidant activity within its cells.After that,the detection of three kinds of kits,SOD,CAT and GSH-PX,showed that licochalcone A can influence the secretion of three kinds of oxidase,and then enhance the antioxidant function of cells.Indeed,oxidative stress can result in cell death due to several different mechanism,including increased formation of reactive oxygen species?ROS?,mitochondrial dysfunction,or activation of apoptosis-related death signaling.Intracellular antioxidant mechanisms involve antioxidant enzymes,including SOD,CAT,and GPx.Moreover,SOD,CAT and GPx serve as the first line of intracellular defense against ROS by catalyzing their conversion to less reactive species.Intracellular ROS levels are tightly controlled by free radical scavengers and by a sophisticated cellular antioxidant system,including SOD,CAT and GPx1 under the action of licochalcone A.Because cell oxidation can lead to canceration of cells,we will further study when licochalcone A enhances the antioxidant capacity of hepatocytes,whether it can also promote the death and apoptosis of hepatoma cells.Finally,by DAPI cell morphology analysis and cell cycle analysis,the final results showed that licochalcone A is safe in the range of application,which can promote the apoptosis of hepatoma cells,and is accomplished through ERK,p38 and JNK pathways in MAPK signaling pathway.As data shown,it was concluded that licochalcone A can enhance the antioxidative property of liver cells and promote the death and apoptosis of hepatocellular carcinoma cells.It has laid the foundation of the development of licochalcone A that is used as the main materials of the natural,safe and efficient health care products.
