| Based on the resources of Hainan Province,the environmentally friendly green chemistry method is used to synthesize hydroxyapatite,a bone material with good biocompatibility,and it is applied as a modifier to other materials.The specific research content is as follows:(1)Firstly,the AHP method was used to optimize the squid bone pretreatment program.The best process for the first soaking sodium hydroxide solution for 12 hours,then saturated sodium chloride solution soaked for 12 hours,and finally soaked in chloroform solvent.Hydroxyapatite(HA)were successfully prepared from cuttlefish bone(CB)powders via hydrothermal solid-state conversion without surfactants or complex-structural agents.The effects of reaction temperature on the microstructure and growth of HA crystals were evaluated by the X-ray diffraction(XRD),and scanning election microscopy(SEM).The Fourier transform infrared spectroscopy(FTIR)revealed that organic matter was detected with no addition of surfactants or complex agents.Thermal analysis(thermogravimetric analysis(TGA)and differential scanning calorimetry(DSC))and X-ray spectrometer(EDS),these samples contain a small portion of aragonite and calcite crystal,which will gradually decrease with increased hydrothermal temperatures.(2)The prepared hydroxyapatite was examined for its biocompatibility and sustained-release properties.The MTT test showed that after seven days of cultivation,MG63 cells covered the surface of the material and accumulated and grew well.In addition,hydroxyapatite prepared using squid bone as a calcium source exhibits more excellent biocompatibility than conventional hydroxyapatite.The drug-loading and sustained-release properties of hydroxyapatite were studied using Hainan specialty drug Naked Flower Purple Pearl as a model drug.The results showed that hydroxyapatite had a low drug loading and poor sustained-release properties.Future solutions need to re-find the model drug or chemically modify its surface.(3)The hydrogel was prepared by blending dopamine-modified hydroxyapatite with gelatin extracted from Hainan tilapia.Scanning results showed that the surface of hydroxyapatite was more uniformly loaded with dopamine oxide.Using the hydroxyl groups contained in polydopamine,horseradish peroxidase as a catalyst,gelatin in addition to its own adhesive,it also weakly bonds with dopaminated hydroxyapatite and forms a water-like structure similar to the interpenetrating network structure.glue.The results showed that the dopaminergic modified hydroxyapatite was homogeneously dispersed in the hydrogel pipeline structure compared to the non-distribution hydroxyapatite,and it played a good role in enhancing the mechanical strength of the hydrogel. |
