Cardiac-cerebral vascular diseases ranks the first place in the disease which causes human death,and hypertension is the most common one.Many available chemical medicines have the curative effect in the treatment of hypertension,but accompanied with many toxic side effects.Semen Raphani is a commonly used traditional medicine in the treatment of digestive system disease,excretory system disease,dermatosis,cardiac-cerebral vascular diseases,cancer,etc.Sinapine thiocyanate,a kind of water-soluable alkaloid,is the key antihypertensive constitute of Semen Raphani.The purpose of this study was to establish the extraction,purification and preparation technology of sinapine thiocyanate from Semen Raphani,to improve the content of effective component in hypotention capsule.The research results are as follows:(1)The HPLC method for determination of sinapine thiocyanate in semen Raphani:Through the investigation of linear relationship,precision,stability,the results showed that the method was reliable,stable and accurate,and could be used for the determination of sinapine thiocyanate in semen Raphani.(2)The extraction conditions for sinapine thiocyanate from semen Raphani were optimized.Water was used as extraction solvent,with ultrasonic power of 400W and solid to liquid ratio of 1:10(w/v).semen Raphani were extracted for 3 times wtih 30 minutes each time.After filtration and concentration,ethanol was added to the concentrate with content of 80%for alcohol precipitation.And then,the supernatant solution was concentrated to removing ethanol.In the whole extraction process,the average transfer rate of sinapine thiocyanate was 82.95%.(3)The optimal macroporous resin purification technology for sinapine thiocyanate from semen RapHani:The elcohol precipitation concentrate was diluted to 0.25g semen Raphani per ml solution(0.25g/ml).The optimal macroporous resins was AB-8,the sample volumn was 3 bed volumm(BV)with flow rate of 1BV/h.After washing with 2BV water,sinapine thiocyanate was desorbed by 4BV of 50%ethanol.The eluant was concentrated and then dried to obtain the resin purification product.The content of sinapine thiocyanate was 27.86%with average transfer rate of 76.02%during the resin purification process.(4)The optimal anti-hypertension capsule preparation technology:Dry resin purification product,microcrystalline cellulose,silica,carboxyl methyl starch sodium with proportion of 40:40:10:10 with 2%poly povidone K30 ethanol(50%)solution preparing ii soft material,preparing granulation in 24 mesh(aperture 850?m),drying at 80?,granulation in 20 mesh(aperture 710?m),and joined the dry particles of 0.5%(w/w)of magnesium stearate as lubricant,uniform mixing,with No.1 capsules filled,The filling amount was about 0.26g per capsule.The operating ambient humidity was below 64.26%.
