Sucralose belongs to the sucrose deoxychlorinated derivative,its sweetness is 600-800 times that of sucrose.It is a functional sweetener,it is not absorbed by the body itself,no calories,similar to the taste and sucrose,high stability,widely used in food and medicine industry.This project uses enzyme-chemical method to design a synthesis of sucralose line,the research includes the following aspects:1.Enzymatic Selective Protection of Sucrose 6-hydroxy is an effective method for the preparation of sucrose-6-acetate.In the DMSO/tert-butanol solvent system,the acyl donor vinyl acetate was esterified with the immobilized lipase Lipozyme TL IM to produce sucrose-6-acetate.The optimal reaction conditions were as follows:V(DMSO):V(tert-butanol)=1:4,sucrose concentration 40g/L,enzyme concentration 100mg/mL,vinyl acetate to sucrose molar ratio 5:1,the reaction time is 24h,the reaction temperature is 30?.2.The 4-OH,1'-OH,6'-OH of sucrose-6-acetate is chemically substituted by a chlorine atom.The Vilsmeier reagent prepared from DMF and solid pH osgene is used to selectively chlorinate sucrose-6-acetate,which has high yield and low pollution.The optimal reaction conditions were as follows:V(DMF):V(trichloroethane)=1:4.5,molar ratio of sucrose-6-acetate to pH osgene was 1:4,reaction time was 3.5h,reaction temperature 115?.3.Enzymatic removal of acetyl chloride sucralose-6-acetate to give sucralose.In t-butanol solvent,the lipase from Aspergillus oryzae is used for deacetylation with sucralose-6-acetate.The method is easy to operate and the product has high purity.The optimal reaction conditions were as follows:enzyme concentration 7mg/mL,V(tert-butanol):V(water)=10:1,sucralose 6-acetate 50g/L,reaction temperature 40?,The time is 6h.4.The TLC analysis and liquid chromatograpH y were used to detect the reaction of sucralose synthesis.The structure of the reactants was determined by melting point determination and IR spectroscopy.Experimental products,after simple processing,the purity of the product was obtained close to the standard crystal samples.
