| Pectin is a heterosaccharide macromolecule existing in plant cell wall.The pectin absorbed after ingestion can not be digested by enzymes in thestomach of single stomach,and is directly hydrolytic by anaerobicbacteria to produce Pectin oligosaccharides(POS).Functional oligosaccharides can activate intestinal probiotics and promote their growth and reproduction.Intestinal bacteria,the bacteria will gradually reduce with age increased.The number of probiotics in the intestinal tract is one of the indicators to measure the health of the human body.With the rapid development of medical science,broad-spectrum and powerful antibiotics are widely used in the treatment of various diseases,so that the balance of normal bacteria in human intestinal tract is damaged to varying degrees.Therefore,to increase the number of intestinal bacteria is very necessary.The intake of probiotic products is simple and reliable,but these products are limited by many conditions from production to sale,and it is more feasible to promote Bifidobacterium natural multiplication in the intestinal tract by ingestion of functional oligosaccharides.The main results of this paper are as follows:1.The effect of alkali deesterification on the quality of pectin was emphatically discussed.The effect of p H and deesterification time on the esterification degree of pectin products was discussed with high ester pectin as raw material and the reaction temperature of 5.The results show that the best process conditions for deesterification of high ester pectin base are as follows: p H is 11.5,deesterification time is 30 min,the reaction is under low temperature,and the index satisfies the standard of the degree of esterification of low ester pectin(DE < 50%).2.Pectin was degraded under the action of pectinase,and many galacturonic monosaccharides could be obtained after the reaction under strong acid conditions.The supernatant was centrifuged to be a mixture of monosaccharides and oligosaccharides.In this paper,the effects of reaction time,reaction temperature,concentration of substrate,concentration of enzyme and p H on reaction were studied in the preparation of pectin oligosaccharides.The optimum reaction conditions were obtained by optimizing the response surface.Pectin oligosaccharides were filtered with 45 microporous membrane.The molecular weight 10 KDa of the membrane assembly was installed.Under the pressure of 0.020 Mpa,the temperature of the pectin solution was 35,and the pectin concentration was 1g/L ultrafiltration,the pectin solution was obtained through the ultrafiltration membrane pressure,the component 1(molecular weight < 3k Da),the fraction 2(molecular weight 3k Da to 5k Da)and the component 3(molecular weight).The amount of 5k Da to l0 k Da).3.The effect of pectin oligosaccharide on Bifidobacterium growth was studied and its fermentation properties were studied.The fermentation broth p H at different time points was determined,and the PI value of the probiotic index was calculated from OD value.The results showed that pectin and pectin oligosaccharides could make Bifidobacterium proliferate,and the probiotic activity of low ester pectin was better than that of high ester pectin,and the pectin oligosaccharide was better than pectin,and the smaller the molecular weight,the better the effect of Bifidobacterium.4.Preparation of pectin oligosaccharide microcapsules: with pectin oligomer as a part of microcapsule wall material,Bifidobacterium microcapsule was prepared with Bifidobacterium as core material,and the best composition of preparation of Bifidobacterium microcapsule wall material was preliminarily determined.Moreover,the number of viable bifidobacteria was observed in simulated gastric juice,different concentrations of bile solution and intestinal fluid.The experimental results showed that the number of living bacteria was still detected after the presence of 3H in the simulated gastric juice.In the simulated bile salt solution,the bile salt with a concentration of 1g/L had no significant effect on the number of living bacteria.The number of living bacteria decreased with the concentration of 3g/L,but the number of living bacteria still existed;the microcapsules were broken and released double in the simulated intestinal liquid.Sod.It shows that the microcapsules prepared by experiments are enteric microcapsules and are beneficial for human health. |
