Raspberry is a deciduous perennial shrub in the genus Rubus L of the Rosaceae family.With various colors,raspberry fruits are tender and juicy,abundant in a variety of physiologically active ingredients,such as vitamins,ellagic acid,raspberry ketones and anthocyanins,thus have the potential for various developments in future.In this study,the extraction conditions of raspberry anthocyanins were systematically screened with raspberry fruits as material,and the conditions of anthocyanins detection in raspberry fruits by UHPLC-MS/MS were further explored,then anthocyanins in raspberry fruits analyzed qualitatively and quantitatively in the aim of providing a technical support to the extraction,development and utilization of the anthocyanins in raspberry fruits.The results are showed as follows:?1?The anthocyanins were extracted from the raspberry fruits by solvent extraction and the content of anthocyanins extracted was considered as an indicator.On the basis of single factor tests,orthogonal test were employed to study the conditions in this experiment.Factors affecting the anthocyanins extraction quantity including the hydrochloric acid volume fraction,extraction temperature,extraction time and solid-liquid ratio were investigated.The optimum conditions of anthocyanins extraction were obtained as:1.5%hydrochloric acid methanol solution as extraction agent,solid-liquid ratio of 1:30?g/mL?,extraction time of 3 h,extracted three times temperature of35?.The content of anthocyanins in raspberry determinated by pH different method was 4.49mg/g.?2?The optimum conditions for the detection of anthocyanins in raspberry were obtained by UHPLC-MS/MS:Chromatographic column ACQUITY UHPLC BEH C18 chromatographic column?100 mm×2.1 mm,1.7?m?;Column temperature is 30?;Sample volume is 5?L;The mobile phase A is ultrapure water containing 0.1%formic acid,and mobile phase B is acetonitrile;The flow rate is 0.2 mL/min;Detector wavelength is 512 nm;Detector time is 30 min.Gradient elution conditions:0-5 min,5%-10%B;5-10 min,10%-15%B;10-15 min,15%-95%B;15-20 min,95%B;20-20.1min,95%-5%B;20.1-30min,5%B.?3?According to the experiment,qualitative analysis shows that 6 kinds of anthocyani ns detected in raspberry fruits.They are Cyanidin-3-O-sophoroside,Cyanidin-3-O-sambubios ide,Cyanidin-3-O-glucoside,Cyanidin-3-O-rutinoside,Pelargonidin-3-O-glucoside and Pelargo nidin-3-O-rutinoside.Quantified by the the standard of malvidin-3-O-glucoside.Quantitative analysis showed that the content of Cyanidin-3-O-glucoside was highest,secondly were C yanidin-3-O-glycoside and Cyanidin-3-O-rutinoside.The study showed that anthocyanins in ra spberry fruits is mainly cyanidin,only containing a small amount of pelargoidin.
