Yangwei granule is a kind of Chinese medicine granules.The main components are Glycyrrhiza,black plum,yam and Paeonia lactiflora,which have the function of nourishing the stomach,strengthening spleen,regulating qi and in the middle,which is one of its main active ingredients.At present,in the process of production,there are some problems such as poor visualization,lack of understanding of the process and effective monitoring means,which lead to the large difference in product quality and unclear attributes,which affect the consistency of product quality.In this regard,the extraction rate of paeoniflorin,an effective active ingredient,was improved by precise control of the ext:raction process parameters of Yangwei granule,and the loss rate of paeoniflorin in two heating processes in the process of granulating was studied,and the overall quality of Yangwei granule from medicinal materials to intermediate to finished product was controlled.The main contents are as follows:1.In order to improve the extraction rate of paeoniflorin in Paeonia lactiflora pall,the parameters of the extraction process were optimized by orthogonal experiment.The results showed that the extraction time was the first 2h and second times 1.5h respectively when the water temperature was 30.8?,two times extraction and 3.5 times water,and the heat preservation temperature was 93 ?.The extraction rate of paeoniflorin was the highest in the decoction.2.A methodological study on the determination of paeoniflorin in Yangwei granule by HPLC(high performance liquid chromatography)was carried out.The results showed that the sample prepared with 50%ethanol to 25ml volume bottle was the best,and the specificity of negative interference showed that the sample of Paeonia lactum was not interfered;the chromatographic peak of paeoniflorin was not detected.Impurities;the relative standard deviation of instrument precision is RSD = 0.26%(n = 6);the relative standard deviation of sample repeatability test is RSD=0.4%(n=6).3.In order to retain the content of paeoniflorin in the production granulating process,the influence of the parameters of the granulating process,such as the content of the extract,the content of paeoniflorin,the vacuum degree,the temperature and the drying time,was studied and analyzed.The results showed that the loss of paeoniflorin content was related to the content of paeoniflorin in the extract and the content of paeoniflorin in the extract during the heating and drying process,and the correlation between the content of paeoniflorin and the loss rate was not obvious in the condition of less water,and the specific gravity of the extract diluent(20?)was measured.The content of paeoniflorin in Yangwei granule can be quickly detected by near infrared spectroscopy,and it can be used to monitor the content of semi finished products and the quality stability.4.The stability of paeoniflorin in the production of Yangwei granules was studied.The results showed that the temperature has the greatest influence on the content of paeoniflorin in the stomach intermediate.When the temperature is below 70?,the loss rate of paeoniflorin content is the lowest in the water decoction.The higher the content of glycosides is,the higher the content loss rate is,the positive correlation exists between them;the content of paeoniflorin in Yangwei granules is not related to the loss rate.To sum up,the content of paeoniflorin in the production process of Yangwei granule is studied in this paper.Through the change and fluctuation of the key content of paeoniflorin in the process of extraction and preparation,the whole production process of Yangwei granule is monitored,the quality index of the intermediate is increased,the process control is optimized and the product is improved.Quality.
