| The whole plant of A.trifoliata has medicinal activity and is a plant with a wide range of medicinal properties.The fruit of Akebia is also a healthy fruit with rich nutrient elements.At present,the research on A.trifoliate mainly focuses on germplasm resources investigation,genetic sequence analysis and fruit peel processing.However,there are few studies on A.trifoliatum seed oil.In this paper,the extraction technology,physicochemical properties,fatty acid composition,active components of seed oil and anti-oxidant anti-tumor cell proliferation activity of Akebia trifoliate seed oil were studied and analyzed.The conclusions are as follows:1.Using Sophora chinensis seed oil extraction rate(mg/g)as an index,the Soxhlet extraction method was compared with Soxhlet extraction,ultrasonication,and ultrasonic assisted water enzymatic extraction,good.Based on the single factor test of extraction temperature(?),extraction time(h)and solid-liquid ratio(g/mL),the orthogonal test was used to optimize the optimal Soxhlet extraction conditions and the best results of the orthogonal test were performed.The results showed that the optimum conditions for Soxhlet extraction were as follows:extraction temperature 40?,extraction time 4 h,solid-liquid ratio(g/mL)1:50.Under these optimal conditions,the seeds of Hubei,Yunnan,and Mayang trifoliate oil were extracted.The acquisition rates were 34.1 ±3.11,33.3±4.12 and 31.45±4.23mg/g,respectively.2.The physicochemical property activity and fatty acid composition of A.trifoliata seed oil extracted under the optimum conditions were measured.The results showed that the three A.trifoliata seed oil had higher quality,with yellow color and Faint scent,no precipitation occurred.The relative density of the three seed oils was between 0.922 and 0.931.The smoke point was higher than the first soybean oil standard,and the moisture and volatile matter contents were normal.The saponification values of seed oil from Yunnan,Hunan and Hubei were 214.54,210.41 and 211.39 KOH mg/g,respectively.Among the three producing areas,the seed oil in Hubei had the highest acid value(0.542 mg KOH/g)and iodine value(75.42 g/100 g),and Yunnan and Hunan were 0.512 mg KOH/g(75.33 g/100 g)and 0.498 mg KOH/g(71.65 g/100 g),respectively.The peroxide values of Hunan and Hubei seed oils were the same with 4.52 mmol/kg while 4.13 mmol/kg for Yunnan seed oil.The composition of fatty acids in A.trifolia seed oil was determined by gas phase analysis.The results showed that the main constituents of A.trifolia seed oil were palmitic acid,oleic acid,and linoleic acid.The content of unsaturated fatty acids was the highest in Hunan,accounting for 74.68%,including oil.The highest acid is 48.701%.The highest content of linoleic acid was found in Yunnan Sanyemutong(32.71%).In addition,the three oils also contain a small amount of a-linolenic acid.3.The active ingredients of the three kinds of seed oils were analyzed.The polyphenol content was 53.34±0.14?58.34±0.002 mg GAE/g,while the flavonoid content was 312.37±24.43-739.45±28.84 mg CE/g and the carotenoid content was 0.78±0.03?2.16±0.06 mg/kg.The antioxidant activity of oils in Yunnan was the highest,and the antioxidant activities of DPPH,ABTS and FRAP were 3.44±0.12,5.55±0.12 and 0.25±0.02 ?mol TE/g,respectively.The cell experiments showed that only the seed oil from Yunnan trifoliate had inhibitory effect on HepG2 cells,and the inhibitory rate was about 20%when the concentration was 6 mg/mL. |
