Lutein is a natural pigment that is a key antioxidant for the development of healthy eyes in infants and young children and has positive effects on the visual development of infants and young children.Therefore,the study on lutein in infant formula milk powder has important significance for the healthy growth of infants and young children.The study found that the stability of lutein was affected by many factors.However,the study on the stability of lutein in infant formula milk powder is limited.Therefore,this study systematically studied the stability of lutein in infant formula milk powder.This study has practical significance for the application of lutein in infant formula milk powder,which can provide a sound and full scientific basis for the healthy development of milk powder industry.The topic first explored the stability of lutein,followed by a validation of the method for determining lutein in milk powder,and then investigated the effects of other nutrients in milk powder on the stability of lutein.The stability of lutein during milk processing and reconstitution and storage,the specific contents of the experiment and the corresponding results are as follows:?1?Study on the factors affecting the stability of lutein:The effects of light,temperature,pH,oxidants,antioxidants,and metal ions on the stability of lutein were investigated.Studies have shown that:light will accelerate the destruction of lutein,the retention rate of lutein is only 55.45%after 240h;the stability of lutein decreases with increasing temperature,and the retention rate of lutein is only 6h after 60°C destruction,It is 65.50%;in the neutral and alkaline environment,the retention rate of lutein remains basically unchanged,and in the strong acid environment?pH 14?,the retention rate of lutein decreases greatly?P<0.05?;when H2O2 concentration was 0.80%,the lutein retention rate decreased significantly after 6 h?P<0.05?,and the loss was as high as 70%;when the concentration of VC and VE was 0.10%,the retention rate of lutein was significantly decreased after 12 hours?P<0.05?,which was 73.45%and 83.31%,respectively;with the metal ions Fe3+,Fe2+,Cu2+coexisted 3h,the retention rate of lutein significantly decreased?P<0.05?,respectively 65.12%,59.34%and 45.22%.The above results indicate that light and prolonged high temperatures have a significant destructive effect on lutein.The stability of lutein is poor in strong acid environment.The basic environment has no effect on the stability of lutein.The oxidants and antioxidants have a great influence on the stability of lutein.Fe3+,Fe2+,and Cu2+in metal ions have a great influence on the stability of lutein and have a strong destructive effect.?2?Determination of lutein in infant formula milk powder:This section fine-tuned the detection method of lutein in the national standard and verified the reliability of the method.The chromatographic column was C18 and the mobile phase was methyl tert-butyl ether-methanol?15:85?;column temperature 25°C;flow rate 1.0 mL/min;detection wavelength of 446 nm,the correlation coefficient of the standard curve is 0.9986,a good linear relationship.The recovery rate of lutein was between 98.2%and 102.7%,and the relative standard deviation was between 1.3%and 2.4%.This indicates that the results obtained by this method have high accuracy and precision and can be used in this study infant formula milk powder determination of lutein content.?3?The effect of nutritional supplements on the stability of lutein in infant formula milk powder:According to the results of the first part of the experiment,copper salts,iron salts,VC,and VE have a significant influence on the stability of lutein,therefore,this part of the experiment examined their effect on the stability of lutein in infant formula milk powder.The results showed that the loss rate of lutein was lower than that of the blanks stored at 37°C for 3 months,with 0.25mg/100 g copper salt,3 mg/100 g iron salt,50 mg/100 g VC,and 4 mg/100 g VE,the groups were0.77%,1.02%,1.11%,and 0.91%,respectively,and there was no significant difference from the blank group?P>0.05?.Loss of lutein in milk powder supplemented with 0.60 mg/100 g copper salt,7 mg/100 g iron salt,100 mg/100 g and 200 mg/100 g VC,12 mg/100 g and 18 mg/100 g VE The rate was significantly increased compared with the blank group?P<0.05?,which were 2.19%,2.31%,5.30%and 8.36%,3.30%and 6.50%,respectively.From the above it can be concluded that for copper salt,iron salt,VC,VE,during the storage period,the loss rate of lutein in milk powder increases with the increase of the amount of each nutrient,and the addition of high nutrient content had a significant effect on the stability of lutein in milk powder?P<0.05?,and the low nutrient addition had little effect on the stability of lutein in the milk powder?P>0.05?.?4?Study on the effect of infant formula formula on the stability of lutein:This study investigated the effects of homogenization,high temperature sterilization,concentration,and spray drying on the stability of lutein.The results showed that the average loss rate of lutein in the homogenization process was less than 1.2%,and there was no significant difference in lutein content before and after milk homogenization?P>0.05?;the average loss rate of lutein in the sterilization process for 6.43%,the lutein content was significantly different before and after high-temperature sterilization of the emulsion?P<0.05?;the average loss rate of lutein in the spray-drying process was 7.49%,and the lutein content in the milk powder was significantly reduced before and after spray drying?P<0.05?;In the concentration step,the average loss rate of lutein was very small at 0.03%,and the concentration process had little effect on the stability of lutein?P>0.05?.It can be seen from the above that during the processing of infant milk formula,the greatest impact on the stability of lutein is the spray drying and high temperature sterilization process.The relatively small impact is the homogenization process and the concentration process.How to improve infant formula milk powder processing technology to improve the stability of lutein in the process requires further in-depth research.?5?The stability and degradation kinetics of lutein during the preparation and storage of infant formula milk powder:This study first studied the stability and thermal degradation kinetics of lutein during milk powder preparation.The results showed that:in milk powder reconstitution,the thermal degradation of lutein is a first-order reaction,and the lutein is at 40°C,50°C,60°C and70°C.The higher the brewing temperature,the faster the degradation reaction and the shorter the half-life.The activation energy Ea of the degradation reaction was 27.62 kJ/mol.After that,several factors affecting temperature,packaging materials,air,and light were selected in the experiment.The stability of lutein in infant formula milk powder storage was studied.It was found that with the increase of storage temperature,three different kinds of packaging materials were used.The retention rate of lutein in milk powder showed a downward trend.Under the condition of 40°C,the lutein retention rate in tinplate cans,aluminum foil PE flexible packaging,and paper cans were significantly lower than those at 10°C and 25°C?P<0.05?,which were 56.88%,61.54%,64.19%,respectively,at 6 months,the internal loss rate is about 40%;and at 40°C,there is a significant difference in the retention rate of lutein between the powdered milk powder of the tinplate cans and the other two sets of powdered milk?P<0.05?;it is also stored at room temperature 25°C for one month.There was a significant difference?15.43%?in lutein retention rate between sealed and exposed environment?P<0.05?.The retention rate of lutein in milk powder stored at 25°C decreased to about 72.20%,and the loss rate under dark conditions was not significant.The difference was significant?P<0.05?.From the above,it can be seen that in order to reduce the loss of lutein,the temperature should be 40°C when formulating milk powder,and it should be consumed as soon as possible after reconstitution.In the storage of milk powder,different temperatures,packaging materials,air and light conditions can cause a large loss of lutein.Under high temperature conditions,lutein in milk powder of different packaging materials was destroyed to a large extent,and tinplate cans milk powder had the greatest effect on the stability of lutein at high temperatures.Therefore,high-temperature light should be avoided during the storage of milk powder,and it should be consumed as soon as possible after opening to reduce air-induced degradation of lutein.
