Exosomes,a double-layer membrane vesicle-like structure with a diameter of about 30-100 nm,can be actively secreted by a variety of cells in the body and widely distributed in various body fluids.Due to its particular structure and function,exosomes have demonstrated great potential in basic research,clinical diagnosis and treatment.Therefore,it is very important to develop an efficient and facile method for exsome isolation.The selectivity of traditional method,such as centrifugation and precipitation was bad,while the cost of immunoseparation method was high.Molecular imprinting technology is a highly selective separation technique emerging at the end of the 20th century.Its basic idea is based on the recognition of antibody-antigen specificity.With extremely high selectivity and excellent molecular recognition performance,the prepared material has been used widely in many aspects.However,the molecular imprinting technology has not been applied to the separation of exosomes.This paper mainly carried out the following research:(1)Based on the combination of aptamer technology and imprinted polymer technology,protein molecularly imprinted materials for lysozyme was prepared.The different and excellent performance were demonstrated after the study of adsorption properties.(2)Tumor-derived exosome molecularly imprinted materials based on the combination of aptamer and nucleic acid amplification was prepared for efficient isolation of exosomes.(3)Integrated with microfluidic chip,the innovative material was used to capture cancer cell-derived exosomes from blood samples of cancer patients.And after the analysis of the contained mutant nucleic acid fragments of tumor cell-derived exosomes,this technique will lay the foundation for the construction of an exosomal-based microfluidic chip cancer liquid biopsy in vitro diagnostic system.
