Slab gel electrophoresis is traditionally used for analytical separation of macro biomolecues.In this form of electrophoresis,staining is normally necessary to visualize the separation results,which is tedious,laborious and time consuming,and online detection is not available.Capillary gel electrophoresis appeared later,which is fast and consumes less sample.Photometric and fluorescence are used for online detection in capillary electrophoresis for solutes with photoactivity.However,labeling is still necessary for online detection of non-photoactive species.Aiming at gel electrophoresis separation of biomolecules and online detection without labeling,we designed a simple and convenient column gel electrophoresis system with coupled voltage detection,and demonstrated the feasibility of gel electrophoresis separation with online detection using small ions.In the first chapter,methods of gel electrophoresis of biomolecules and capillary electrophoresis of inorganic ions are summarized.The principle of conductivity detection and methods of simultaneous separation of detection of cations and anions are introduced,In the second chapter,column gel electrophoresis of inorganic ions and online detection with coupled voltage is presented.NH4+,K+,Na+,Li+,Mg2+,F",Cl-,SO42-are successfully separated and detected in a histidine buffer system.The applicability is demonstrated by ion analysis of beverages.Simultaneous analysis of cations and anions with one time sample introduction at the middle of the column is also realized and used in the analysis of ions from serum sample.In the third chapter,separation and online detection of two model amino acids without labeling are demonstrated.In 2.3 mol/L acetic acid system,L-histidine and glycine are well separated and detected,demonstrating the potentials of the method in the separation and non labeling online detection of biomolecules.The method present in this work is simple,portable and easy to operate,and suitable for field analysis with further system miniaturization.
