Abalone is an ancient sea creatures.Most people like it very much all over the world,because of the tender smooth,the fresh and not greasy meat.Abalone is the most famous seafood compared to ginseng,wings and belly.During Ming-Qing Dynasty,abalone was regarded as one kind of "sea Bazhens".It is not only a kind of expensive food,but also a kind of medicine.However,abalone viscera,30%?40%of the abalone gross weigh are discarded during the processing.Just few was used to process as animal feed,resulting in a waste of resources.It contains a lot of proteins,vitamins,bioactive substances and other nutrients.Our objective was to use the connective tissue of abalone viscera as raw material,to prepare the collagen ACE inhibitory polypeptides from it by separation and purification methods.The molecular weight of the collagen was also measured in the article.(1)The effect of separation factor and the molecular weight of the ultrafiltration membrane on ACE inhibition rate of collagen peptides were determined by single factor experiment.The results showed that inhibition rate of ACE collagen peptide was 32.1%.It could effectively remove the pigment,proteins and macromolecules of sample at 35185.Considered from the aspect of energy consumption and others,the optimal effect of ACE inhibition rate of collagen peptides was 40.9%when the molecular weight of ultrafiltration membrane was 3KDa.(2)In the process of gel chromatography of collagen peptides,the effects of eluent flow rate,eluent concentration and upper concentration on ACE inhibition rate were studied by single factor and orthogonal experiment.The results showed that the order of influencing factors was as follows:eluent concentration>sample concentration>eluent flow rate.The upper concentration and eluent concentration had significantly impact on the ACE inhibition rate of abalone organs collagen.However,the eluent flow rate had insignificant effect on it.Thus,the optimum conditions for gel filtration chromatography were:sample concentration(60mg/mL),eluent concentration(0.1mol/L)and eluent flow rate(1.0mL/min).(3)The effects of duration of dialysis,dialysis temperature and the number of the dialysis on the desalination rate was researched by single factor experiments in the process of separation and purification of abalone viscera collagen peptides.The results showed that each factor all had significant effect on desalination rate of collagen peptides in the purification process.(4)On the basis of single factor,the dialysis time x1,dialysis temperature x2 and the number of dialysis x3 were studied by quadratic general rotation design,for the optimal test of the ACE inhibition rate of abalone viscera collagen peptides.Regression equations abalone viscera collagen peptides desalination were established;analysis of variance for the regression equation fit for significance testing.The regression equation desalination rate was:Y = 87.16 + 3.08 x1 + 6.67 x2 + 5.17 x3 +0.77 x1 x2-1.78 x1 x3-1.68 x2 x3-2.39x,2-8.80 x22-3.53 x32.Synergistic effect of each index was analyzed and the index of desalination rate was optimized.The optimum process about desalination of collagen peptides obtaining the highest desalination rate(88.24%)were dialysis time(8h),dialysis temperature(36.62?)and the number of dialysis(4 times).It was similar to the forecasting of optimization group(88.42%),which proved the reliability and validity of quadratic regression equation.(5)Determination of molecular weight of abalone viscera collagen peptide:The sample was mainly in the molecular weight of 1216.56Da?911.47Da?1340.67Da,that was mainly 8 to 12 amino acid residues of the peptide.
