Yeast haploid and diploid are widely applied both in production and scientific research.Transformation between haploid and diploid,and their identification are great significance to manufacture and life science.Inducing yeast sporulationand and then obtaining haploids are fantastically crucial for yeast genetics research and breeding,menwhile,it is necessary for character genetic analysis,but also making a good prepare for hybriding.This experiment conducted S.cerevisiae haploid screening and fermentation properties measurement,hoping to gain haploids with excellent fermentation characteristics or extreme phenotype,and to provide hybrid material for subsequent QTL mapping.The results were as following:1.In 12 diploids that were observed spores,as time extened,the spore production rate rised to stabilize after a fixed point without continuing to rise.In terms of spore production rate,spore production stable time,ascus types,and genotype,LFN520,44y7,112y4,112y14,174y1,31y3,31y10 and 33y7 were selected to induce sporulation and haploid separation.2.After chromogenic reaction,34 haploids with extremely H2S-producing phenotypic were acquired from 106 haploids,containing 13 low-H2S production haploids;91 haploids can be seen as extremely aerosis phenotype haploids;29 haploids can be seen as extreme flocculation phenotypic strains.3.31y3-33,31y10-19,44y7-1 were high-glycerol-production haploids with respectively 7.63 g/L,8.19 g/L,7.40 g/L,and 174y1-3,174y1-5,174y1-6,174y1-27 were low-glycerol-production haploids with glycerol production respectively 4.33 g/L,4.60 g/L,4.55 g/L,4.71 g/L;33y7-43(11746.20 mg/L),112y4-45(986.88 mg/L)can be considered as the extremely high-higher alcohols-production phenotypicstrains,otherwise,31y3-33(127.43 mg/L),44y7-1(178.77 mg/L),174y1-5(151.65 mg/L)can be seen as extremely low-higher alcohols-production phenotypic strains;174y1-3(75.78 mg/L),112y4-45(42.90 mg/L)can be regarded as extremely high-esters-production phenotypic strains,meanwhile,31 y3-3 3(24.41 mg/L),112y4-46(27.75 mg/L),44yy7-1(24.67 mg/L),174y1-5(27.69 mg/L)can be seen as extremely low-esters-production phenotypic strains.4.From respect of choosing optimum fermentation characteristics strains,haploids 112y4-45,112y4-46,174y1-5 had significant advantagees;From the angle of choosing extreme phenotype strains,31y10-19 and 174y1-3,33y7-43 and 174y1-5 had extremely opposite fermentation characteristics.Despite of opposite mating type,it will be changed by knocking out to hybride,providing materials for QTL mapping.
