Because of the special natural environment in Lanzhou,the uniqueedible lilium varieties were raised,which is a famous specialty of Gansu Province,the colorwhite as jade,fleshy sweet.Annual production of fresh Lilium davidii var.unicolor Salisb in Lanzhou reach up to more than 100,000 tons,the output value is more than 50 million yuan is one of the important pillar industries.But Lilium davidii var.unicolor Salisb in storage and processing is difficult to guarantee the quality and shelf-life,and the bulb will produce a lot of waste,so this paper development of dried Lilium davidii var.unicolor Salisb non-sulfur anti-browning and use the generated early in the process of waste bulbs to extraction of polysaccharide after separation and purification,sulfated modification,and study the in vitro antioxidant activity.Mainly contains the following aspects:(1)Discusses the ascorbic acid,citric acid,sodium chloride andpolyphenols of non-sulfur anti-browning effect,and the effect of gradient temperature,constant temperature continuous,gradient cooling of three hot air drying methods on the color protecting Lilium davidii var.unicolor Salisb stem product quality.The results showed that the composite color fixative optimum combination of ascorbic acid0.04g/100mL,citric acid 0.3g/100mL,sodium chloride 0.3g/100mL,soak60min,color protecting effect is the best;three hot-air drying method,using gradient temperature dry Lilium davidii var.unicolor Salisb rehydration best,browning at least.(2)In this study,ultrasonic-assisted compound enzyme hydrolysis extraction and microwave-assisted extraction methods were employed to separate polysaccharides from Lilium davidii var.unicolor Salisb to simulate industrial production and the process parameters were optimized(The optimal parameters of ultrasonic-assisted compound enzyme hydrolysis extraction of polysaccharides from Lilium davidii var.unicolor Salisb(LPS-CE):rennet enzyme dosage 1.5%,fiber luciferase enzyme dosage2.0%;ultrasonic temperature 50℃,extraction time 25min,solid-liquid ratio 1:25,ultrasonic power 225W.The optimal extraction process of microwave-assisted(LPS-M)is:microwave power 597W,extraction time 60min,solid-liquid ratio 1:65,extraction temperature 50℃).In this condition,the extraction yields were 39.86%,36.55%,the total sugar contents were 94.23%,92.85%.Monosaccharide composition was determined by GC-MS method.LPS-CE and LPS-M was mainly composed of glucose and mannose,with the molar ratio of 4.7:5.2 and 5.17:4.82.The weight average molar mass(MW)determined by SEC-LSS were 1.195×105and 1.186×105.The results showed that the extraction yields,content of carbohydrate,monosaccharide composition and weight average molar mass of ultrasonic-assisted compound enzyme hydrolysis method and microwave-assistant extraction method were slightly different.Howerever,extraction time and material-water ratio with ultrasonic-assisted compound enzyme hydrolysis were less than microwave-assistant extraction method.(3)We Obtained sulfate polysaccharide(SLPS-CE)was achieved with the optimum condition(reaction time was 1.5 hours,reaction temperature was 43℃and volume ratio of CSA to Py was 1:1,under these conditions the sulfur content in SLPS-CE can be up to 10.98%+0.216%).The resulted SLPS-CE was characterized by FT-IR.The peaks at 813.55cm-1,1236.58cm-11 indicated that SLPS-CE possesses typical absorption peak of sulfated polysaccharides.Monosaccharide composition of SLPS-CE determined by GC-MS were lower ratio of mannose and higher ratio of glucose because of sulfated modification.The result,determined by SEC-LSS indicated the chain of sulfated polysaccharide was degradation by sulfated reaction.(4)Anti-oxidant activity with effect of scavenging DPPH radicals,OH radicals and superoxide radicals and reducing power and chelating effect on ferrous ions was studied on LPS-CE、LPS-C、LPS-M and SLPS-CE with different extraction method.The result showed that anti-oxidant of LPS-CE was slightly higher than LPS-C and LPS-M,anti-oxidant of SLPS-CE were significantly higher than LPS-CE,Based on experiments,SLPS-CE can be a new source of natural antioxidants with potential value in health food.
