Capillary Electrophoresis-Electrochemiluminescence has become an important and powerful detection method due to the advantages:short analytical time,high efficiency,minimum consumption of reagents of CE,and wide linear range,high selectivity,good stability of ECL.nowadays,the CE-ECL based on the Ru(bpy)32+ system has been extensively applied in diverse areas of analytical chemistry,e.g.,clinical and pharmaceutical assays,food and environmental applications,and thermodynamic and kinetic studies.This thesis is concentrated on the application of EC-ECL for the analysis of Psychotropic drugs and Lycoris Radiata.The main contents and results were introduced as follows:1.Based on electrochemiluminescence(ECL)intensity of ruthenium(?)tris(bipyridine)at a platinum electrode was greatly enhanced by Chlorprothixene,the simultaneous determination of Chlorprothixene in mouse plasma has been developed using capillary electrophoresis with ECL.Under the optimal conditions,the linear ranges over the from 0.0005 to 2 ?g/mL with a detection limit of 0.17 ng/mL(?=3).The relative standard deviation of the peak height was 1.1%(n=6).The recovery in mouse plasma was between 98.1%and 98.5%.2.A novel method for the determination of Tazettine in the bulb of Lycoris radiata was proposed by using capillary electrophoresis coupled with electrochemiluminescence.The calibration curve was linear over the range from 0.001 to 5.0 ?g/mL with a detection limit of 0.038 ng/mL(?=3).The relative standard deviation of the peak height was 1.8%(n=6).The recovery in the bulb of Lycoris radiata was between 95.1%and 97.1%.3.Based on the enhancement of tris(2,2-bipyridyl)ruthenium(II)with Homolycorine and Lycornine,a novel method for the determination of Homolycorine and Lycornine in the bulb of Lycoris radiata using capillary electrophoresis coupled with electrochemiluminescence has been developed.The optimized detection conditions were 1.2 V potential,5.0 mmol/L Ru(bpy)32+ 50 mmol/L pH 9.0 PBS in detection cell;separation buffer for 10 mmol/L PBS with pH 5.5;separation voltage for 12 kV;injection voltage for 14 kV and injection time for 10 s.Under the optimal conditions,the linear ranges were from 0.2 to 10.0 ?g/mL for Homolycorine with a detection limit of 0.068 ?g/mL(?=3),and from 0.01 to 5.0 ?g/mL for Lycornine with a detection limit of 0.0046 ?g/mL(?=3).This method was successfully applied to detect Homolycorine and Lycornine in the bulb of Lycoris radiata,and the recoveries were between 94.2%and 97.6%.
