Study On Antioxidant Activity Of Red Pigment From Jujube Peel In Vitro

Jujube as a medicine and edible food,loved by the people.Red pigment extracted from it is a natural plant pigment resources,bright color,rich and pure,good stability,good water solubility,safety and health,widely used,prospects.In addition,had rich antioxidant components,mainly containing flavonoids,polyphenols,antioxidant active substances polysaccharides and vitamins,etc.,had a good antioxidant.In this paper,peal waste of Ling-wu long jujube as raw material,red pigment extracted from it.Determined the content of its antioxidant components and research jujube red pigment in vitro antioxidant activity and analyze the correlation with antioxidant activity.Establish H202 oxidative stress model to study the dates of red pigment to improve the role of oxidative stress,cell damage and dose-response relationship.Mainly in the following conclusions:(1)Mainly measured antioxidant active substances on alkaline extraction,ultrasound-assisted alkaline extraction,enzyme-ultrasound-assisted alkali extraction.Pigment alkaline extraction of total polyphenols,total anthocyanins and total flavonoids were the highest,respectively 12.580�0.042 mg/g,0.160�0.02 mg/g and 3.200�0.007 mg/g,ultrasound-assisted alkali extraction of total polyphenols the total anthocyanins and total flavonoids was 9.045 �0.001 mg/g,0.022�0.007 mg/g and 0.368�007 mg/g,enzyme-ultrasound-assisted alkali extraction of total polypheno ls,total anthocyanins and total flavonoids were 6.325�0.013 mg/g,0.124�0.003 mg/g and 0.5227�0.001 mg/g.Different ways of jujube extract red pigment on DPPH�,�OH,I2-�,NO2-�,ABTS+�,H2O2 has some scavenging activity,alkaline extraction were better than the others,significant difference(P<0.05).Macroporous resin purification of the pigment was tested too,the difference between the antioxidant capacity of purification before and after analysis showed no significant difference between them(P>0.05),so the choice was purified alkaline extraction pigment basis to provide raw materials for further research and technical support..(2)The dates of red pigment to change various aspects of cell ultrastructure with improved effects,can significantly improve the damage to endothelial cells,from nuclear to have played a significant qualitative change.The overall shape of cells gradually become the rule,the nucleus shape becomes more and more rules,two-tier structure gradually became apparent nucleolus began to clear,euchromatin gradually increased with increasing concentration,relative heterochromatin less cytoplasm It organelles dye concentration increases gradually become rich,mitochondria,ribosomes,the number of Golgi and endoplasmic reticuhum,the extent of rules gradually to normal cell development.Dates before adding red pigment and then H2O2 treated cells repair the best performance in the nucleus of an irregular situation abound,fuzzy two-tier structure of the nuclear membrane,the effect is not good repair effect of the first two.Jujube red pigment can significantly reduce the level of stress within the cell ROS ease the antioxidant system damage,to further reduce the extracellular LDH activity slows cell damage and further reduce the inflammatory cytokines TNF-a and adhesion molecules ICAM-1,E-Selectin release,avoid further damage caused by inflammatory cytokines,but also to improve the intracellular energy sources mitochondrial function,alleviate lower membrane potential under stress,mitochondrial damage reduction,so as to protect cells.
