Lactic acid bacteria can regulate the balance of ecological flora in human or animal intestinal environment,improve the immune protection ability of the body,promote the repair of inflammation,improve the nutritional metabolism of the body,promote the transformation and absorption of nutrients,and improve food flavor.As a health food additive,the development of lactic acid bacteria has been widely concerned.However,the source and biological characteristics of lactic acid bacteria have an important influence on the effect of lactic acid bacteria.In order to lay a foundation for the development and application of health probiotics germplasm resources,lactic acid bacteria with good anti heterotrophic performance and good effect on intestinal flora and immune function were screened from the feces of healthy infants.The results are as follows:1.Three strains of lactic acid bacteria(le-3,LE-7,le-9)were isolated from feces of healthy infants by routine isolation and identification of lactic acid bacteria and 16 S r DNA molecular biological detection technology.2.Under pH 3.0,the survival rates of E.faecalis le-3,LE-7 and le-9 were 80.57%,84.73% and 87.67% respectively;after 0.3% bile salt treatment,the survival rate was still more than 60%;all the three strains of E.faecalis could inhibit the growth of E.coli,but only LE-7 could inhibit Salmonella,and the diameter of inhibition zone was 14.43mm;none of them had antibacterial effect on Staphylococcus aureus.3.The optimal culture conditions for the three strains of Enterococcus faecalis were as follows: Mrs liquid medium was used as the basic medium,dextrin,beef extract and sweet potato juice were used as the carbon source,nitrogen source and enrichment substance respectively,and then they were cultured by box Behnken The optimal fermentation results of le-3,LE-7 and le-9 were 3.5%,3.5%,3.6%(dextrin),1.8%,1.8%,2.0%(beef extract),12.6%,12.6% and 14%(sweet potato juice),respectively.4.By evaluating the effects of three strains of Enterococcus faecalis on body weight,organ index and intestinal microbial diversity of mice,it was confirmed that the three strains of Lactobacillus could inhibit the growth of harmful bacteria and improve the abundance and species diversity of intestinal beneficial bacteria.5.The three strains of lactobacillus can increase the secretion levels of IgG and SIgA,increase the levels of anti-inflammatory factors(IL-2,IL-4,IFN-?)and reduce the content of pro-inflammatory factors in the intestine of healthy mice,which indicates that the three strains of Enterococcus faecalis can regulate the intestinal immune function of mice.In conclusion,conventional isolation and identification methods and 16 S were used Three strains of Enterococcus faecalis(le-3,LE-7,le-9)isolated from healthy infant feces were screened by r DNA molecular biological detection technology.They have excellent biological characteristics of inhibiting the growth of pathogenic bacteria,adapting to acidic and bile salt environment,regulating intestinal flora and improving intestinal immune function,and optimizing their culture conditions,which provides a theoretical reference for the development and application of human health microecological agents.
