With the rapid development of computer technology,people have made more and more intensive research on micro-nano scale imaging,and micro-nano scale nondestructive imaging often uses optical microscopy,so the requirements for optical microscopes are getting higher and higher.Especially for vivo imaging,it requires not only non-destructive imaging,but also high spatial and temporal resolution.Even though optical super-resolution imaging has been implemented,its temporal resolution is still insufficient.However,the contradiction between temporal resolution and spatial resolution is an important issue that needs to be solved urgently now.How to improve the temporal resolution while ensuring the same high spatial resolution is the current research hotspot.In this paper,we study the resonance scanning technology to improve the temporal resolution Based on the laser spot scanning imaging technology,we design a dual working mode tri-mirror laser scanning confocal microscopy imaging system,and proposes a new galvanometer system structure which is composed of three galvanometer galvanometers and a resonant galvanometer.This structure not only combines the advantages of resonance scanning and galvanometer scanning(high imaging signal-to-noise ratio,high temporal resolution,large field of view,etc.),but also can greatly reduce beam jitter.In addition,the dual working mode tri-mirror laser scanning confocal microscopy imaging system is compatible with four excitation wavelengths,enabling multicolor imaging of cells.Aiming at this system,the imaging control software has been programmed for this system.This software has extremely high flexibility.All microscopy systems based on laser galvanometer scanning can use this software for imaging.Experiments have shown that at a resolution of 512*512 pixels,the frame rate of each wavelength is 45 frame per second,which is 1.45 times the frame rate of ordinary resonance scanning microscopy imaging(The frame rate of ordinary resonance scanning microscopy is only 31-32 frame per second.).
