In this study,the fluorescence photoelectric detection system was studied based on real-time PCR devices with different microfluidic systems.The real-time quantitative PCR system and digital PCR system with special design were studied and tested respectively.Real-time quantitative PCR systems realize temperature cycling based on a Peltier semiconductor,and such systems do not allow continuous shooting.Therefore,from the perspective of optoelectronic control,a photoelectric feedback automatic fluorescence detection system was designed by combining a common PCR instrument with a smartphone/camera.The detection system controls the mobile phone/camera to take pictures by comparing the changes of the selected pixel area on the screen and automatically feeds back.This method greatly improves the accuracy of the shooting time and the clarity of the photograph.This makes the ordinary PCR instrument without fluorescence detection capability acquire the function of real-time fluorescence quantitative PCR3D microfluidic chips and a single heating source can form the temperature conditions required for PCR,which makes the generation of microdroplets independent of expensive chips.Complementary metal oxide semiconductor(CMOS)image sensors are used as detection tools to obtain fluorescent videos of the whole temperature cycle region or the designated cycle region.With video processing software,the amplification information of each stage of the gene can be obtained by monitoring all stages of the PCR globally.Chip digital PCR is the most widely used PCR device at present.The aforementioned 3D microfluidic digital PCR fluorescence detection system was redesigned and optically optimized to suit the low-cost digital PCR chip temperature cycling platform,which resulted in a new chip-based digital PCR fluorescence detection system.Experiments have proved that the system is much more versatile than the commercial digital PCR system in nucleic acid reagents.With the help of the fluorescence detection system of this subject,the above PCR system realized the analysis of inactivated HBV serum samples,and obtained the same results as commercial PCR instrument.While maintaining stable response,the size and cost of the whole system are greatly reduced.
